This study aimed to obtain the right planting system in a greenhouse-scale oriental makuwauri mel... more This study aimed to obtain the right planting system in a greenhouse-scale oriental makuwauri melon plant breeding program and knowing the interaction between the line and the oriental melon growing system at the greenhouse scale. The research design used a Randomized Complete Block Design (RCBD) with two factors, namely melon lines (ougan makuwauri and ginsen makuwauri), and planting systems (tunnel, bracket pot, and vines rope), repeated four times. Observational data were analyzed using analysis of variance and least significant different (LSD) test at 5% levels. The results showed that the best planting system is the bracket pot. This planting system has a unique and attractive appearance as well as more optimal growth compared to tunnel systems and vines rope. Based on the qualitative variable data tested, namely vine length (122.25 cm), stem diameter (0.79 cm), and petiole length (11.86 cm), the bracket pot planting system was significantly different from the tunnel and vines rope planting system. There was an interaction between the lines and the planting system on the observed variables, namely vine length, number of leaves, leaf width, leaf length, petiole length, and stem diameter. This research indicates that bracket pot planting system is recommended for oriental melon plant breeding program.

The lowlands became one agroecology in the chili multi-location test because most farmers, especially in Sumatra, plant chilies on the lowlands after the rice planting season. The purpose of this study was to understand the qualitative and quantitative characteristics of chili genotypes in a lowland area. The experiment was arranged in a single-factor randomized complete block design (RCBD) with three replications. The chili genotypes evaluated were four test lines and six check varieties. The quantitative data were statistically analyzed using analysis of variance (ANOVA) followed by evaluation of genotype means using the least significant difference (LSD) tests by SPSS Statistics software. The results showed that the check variety having the lowest productivity was CH3 (1.19 tons ha -1 ) and the highest was Elegant (4.55 tons ha -1 ). The productivity of four genotypes from IPB were significantly different from the CH3 and Imperial 10 varieties, but not significantly different from Baja (2.96 tons ha -1 ), Balebat (3.87 tons ha -1 ), Elegant (4.55 tons ha -1 ) and Gada (3.04 tons ha -1 ) varieties. Gembili tuber glucomannan contains high fiber. Gembili seeds are becoming scarce, causing gembili plants to be very rarely found in the community. In the West Papua region, local people still rely on conventional supplies which are feared to threaten the gembili plant population. Tissue Culture Technology is very necessary in multiplying gembili seeds quickly and without knowing the season, by growing callus, plant propagation does not require a lot of explants to produce a large number of seeds. The research was conducted at the Politeknik Negeri Lampung Tissue Culture Laboratory, from May to July 2022. The research used a randomized block design, with each treatment repeated three times. The results showed that the composition of the media with the addition of 2.4 D 2 mL.L-1 produces an average of 8.00 days of callus growth, however, for callus growth the culture media with the addition of 2.4 D mL.L-1 produces a value best. 1.73 cm for callus diameter and a value of 0.22 g for callus weight.

After the introduction of plants through the process of adaptation and selection can be used as material crosses with cultivars that have adapted well. A cultivar is said superior when it has a high yield, good fruit quality, resistance to pest attack (plant pests), and able to adapt to various environments grow. The aim of this study was obtain a description, test and compare the yield and quality potential of eight varieties of tomatoes grown in the lowland which are expected to serve as a cross parent material in breeding programs. The experiment was arrange by randomized block design with three replications. The data were analyzed using F-test and the results were significantly midpoint test using LSD test at 5% level. Based on observations on eight strains of local tomatoes and introductions have descripted then selected three strains of tomato introductions which has the form of a round (TM 130804, TM 130805, TM 130806) as well as the introduction of two strains of tomato fruit has an oval shape (TM 130801 and TM 130803) that can be used as breeding material further.

Sweetness level data were significantly different by 9.3 °brix andl0.15 °brix when compared to control (8.9 °brix). Varieties WM 200703 and WM 200706 have bright green skin color with thin stripes with red and yellow flesh color. WM 200704 and WM 200705 have red flesh and dark green skin with thick striations. WM 200703 has the highest fruit weight of 4.63 kg per fruit. The six hybrid varieties tested have the potential to be submitted for registration of the F1 hybrid watermelon variety.

Research result for 7 days of storage the highest germination rate of 10 percent in treatment S1 (2 milligram per litre 6- Furfuryl amino purine (KIN)) which was encapsulated with 2 percent sodium alginate (E1). During the 28 days storage the germination rate increased significantly to 30 percent in the S2 treatment (1 milligram per litre KIN plus 0.5 milligram per litre naphthalene acetic acid (NAA)) and S3 (2 milligram per litre KIN plus 0.5 milligram per litre NAA) which was also encapsulated with E1. Kelompok Tani Multi Baliwo merupakan Kelompok Tani penghasil padi yang berada di Desa Purwokencono, Kecamatan Sekampung Udik, Kabupaten Lampung Timur, Provinsi Lampung. Sehari-hari kelompok Tani Multi Baliwo melakukan kegiatan usaha memproduksi padi yang dibudidayakan secara organik. Masalah yang saat ini dialami oleh Kelompok Tani Multi Baliwo adalah varietas padi Mentik Susu yang mereka tanam secara genetik tidak murni lagi. Masalah ini sulit diselesaikan karena mereka tidak punya pengetahuan dan keterampilan tentang pemurnian genetik benih. Tujuan pengabdian adalah membekali khalayak sasaran dengan pengetahuan dan keterampilan teknik pemurnian benih padi mentik susu yang secara genetik sudah terkontaminasi. Untuk meningkatkan penguasaan materi secara teori, metode yang digunakan adalah melalui ceramah dan memperlihatkan beberapa foto atau gambar kegiatan pemurnian benih padi di lapangan. Untuk memudahkan alih penguasaan keterampilan pemurnian benih di lapangan kepada khalayak sasaran, maka digunakan metode bimbingan teknis dan pembuatan demplot tentang pemurnian benih padi mentik susu di lahan milik petani dan melakukan kunjungan ke lahan Teacing Farm milk Program Studi Teknologi Perbenihan Polinela. Hasil pengabdian menunjukkan semua khalayak sasaran (100%) terjadi perubahan penguasaan pengetahuan dari tidak tahu dan tidak mengerti menjadi tahu dan mengerti tentang cara pemurnian benih padi. Hasil pemantauan lapangan, 100% peserta pengabdian bisa menerapkan teknologi pemurnian benih padi yang secara genetik mengalami kontaminasi.

Therefore, efforts to inventory the local varieties through the establishment of garden collection. As an initial step in the effort to do an inventory of the local varieties of banana varieties exploration contained in Tanggamus district. Exploration banana varieties in the district Tanggamus done in the village of Karang sari in January to March 2014. The research was conducted by means of a survey of local varieties of banana, then the identification of phenotypes and environmental character of the local varieties grown Tanggamus the place of origin is based on interviews of people or farmers around. Then performed field data documentation or passport discriptor accommodate the data, the results of the characterization of the phenotype and the environment. In addition, samples were taken of planting material. Based on the exploration results obtained contained 20 varieties Tanggamus. Varieties of bananas are a favorite in the village of Karangsari varieties of banana juice muli, janten bananas, and bananas. Further planting material in the form of buds planted in polybags as seedlings to be planted in the garden collection.
Program pemuliaan tanaman di kampus Politeknik Negeri Lampung telah memiliki 8 galur semangka gen... more Program pemuliaan tanaman di kampus Politeknik Negeri Lampung telah memiliki 8 galur semangka generasi ke-5 (S5). Galur-galur tersebut telah dievaluasi dan diseleksi dengan harapan telah stabil secara genetik, serta mengetahui level adaptasi terhadap lingkungan. Hasil analisis data menggunakan uji F menunjukkan perbedaan yang nyata antar perlakuan pada tiap parameter pengamatan dengan uji BNT 5%. WM 140502 memiliki berat tertinggi per buah 3.17 kg. Kadar gula tertinggi adalah WM 140507 yaitu 9.47 brix. Saat ini Politeknik Negeri Lampung memiliki 8 galur tipe oval dengan 6 galur berdaging merah dan 2 galur berdaging orange. Dari hasil evaluasi (S5), kita berharap memiliki galur murni untuk perakitan hibrida dengan karakter yang berbeda.
Lipocalins are very important proteins for stress resistance in plants. To better understand the ... more Lipocalins are very important proteins for stress resistance in plants. To better understand the function of tomato lipocalins, we observed responses to oxidative stress using over-expressed SlTIL1, SlTIL2, SlCHL, and silenced-plants. Significant differences in reactive oxygen species accumulation (oxidative damage) were observed in all tested plants under heat stress. Plants with over-expressed SlTIL1, SlTIL2, and SlCHL showed less oxidative damage compared with wild-type plants under heat stress. The expression of SlSODs was induced in over-expressed SlTIL1, SlTIL2, and SlCHL plants under normal and heat stress conditions. Furthermore, silenced PDS, SlTILs, and SlCHL plants showed slightly increasing oxidative damage under heat stress alongside with lower SlSODs under normal and stress conditions. These results suggest that SlTIL1, SlTIL2, and SlCHL were involved in antioxidant defense by eliminating ROS in tomato plants.
In this study, two temperature-induced lipocalin genes SlTIL1 and SlTIL2, and a chloroplastic lip... more In this study, two temperature-induced lipocalin genes SlTIL1 and SlTIL2, and a chloroplastic lipocalin gene SlCHL were isolated from 'Micro-Tom' tomato. The coding sequences of SlTIL1, SlTIL2 and SlCHL were 558, 558, and 1002 bp, respectively. By TargetP analysis, no characteristic transit peptides were predicted in the proteins of SlTIL1 and SlTIL2, while a chloroplastic transit peptide was predicted in the protein of SlCHL. The subcellular localization results indicated that SlTIL1 and SlTIL2 proteins were major localized in the plasma membrane, while SlCHL was localized in chloroplast. To understand the function of lipocalins, transgenic tomato over-expressed SlTIL1, SlTIL2 and SlCHL and their virus-induced gene silencing (VIGS) plants were generated. The phenotypes were significantly affected when the SlTIL1, SlTIL2 and SlCHL were over-expressed or silenced by VIGS, which suggested that the three lipocalins played important roles in regulating the growth and development of tomato. In addition, the level of ROS (O 2 − and H 2 O 2) was low in SlTIL1, SlTIL2 and SlCHL overexpressed plants, while it was high in their silenced plants. The changes in the expression of SODs were consistent with the accumulations of ROS, which indicated that lipocalins might have an important role in abiotic oxidative stress tolerance in tomato plants. Especially SlTIL1 and SlTIL2 are localized around their membranes and protect them from ROS. The results will contribute to elucidating the functions of lipocalin in plants, and provide new strategies to improve the tolerance to abiotic stress in tomato plants.

The experiment was arranged in a Randomized Block Design with 18 two replications. Genotypes tested were introduction and landraces. Phenotypic performance growth type fruit color and productive branch number were realtively uniparm while head of plant, fruit diameter, fruit thick and plant age had were different. Tomato lines FMTT 270, FMTT 22 and FMTT 95 had healthy fruit weight per plant, fruit diameter and good thickness at Lembang highland. Lokereal tomato lines PT 4289, PT 4226, PT 4165, and PT 4121 had earlier harvested, to be further use as breeding material.
