A fundamental aspect of successful social interactions is the ability to accurately infer others' verbal communication, often including information related to the speaker's feelings. Autism spectrum disorder is characterized by language and social-affective impairments, and also aberrant functional neural responses to socially-relevant stimuli. The main objective of the current research was to examine the behavioral and neural effects of making affective inferences from language lacking overt prosody or explicit emotional words in individuals with and without autism. In neurotypical individuals, the current data are consistent with previous studies showing that verbal emotional stimuli enhances activation of brain regions generally responsive to discourse, and also "social-affective" brain regions, specifically medial/orbital frontal regions, bilateral middle temporal areas, temporal parietal junction/superior temporal gyri and pCC/PC. Moreover, these regions respond differentially to positive and negative valence, most clearly in the medial frontal area. Further, results suggest that mentalizing alone does not account for the differences between emotional and neutral stories, as all of our stories required similar inferencing of the feelings of the protagonist. In autism, there is general agreement that the neurodevelopmental disorder is marked by impairments in pragmatic language understandings, emotional processes, and the ability to "mentalize," others' thoughts, intentions and beliefs. However, findings are mixed regarding the precise nature of emotional language understandings. Results of the present study suggest that autistic individuals are able to make language-based emotional inferences, and that like neurotypical controls, social-affective brain regions show task-related facilitation effects for emotional compared to neutral valence. However, the neural activations in the autism group were generally greater than controls, especially in response to emotion. Additionally, results showed greater difficulty with incongruent judgments in participants with autism. Together, these findings represent a first step toward revealing social-affective abilities in the language context in autism, despite irregular brain response. Such understandings are critical to generating effective intervention strategies and therapeutic practices for autistic individuals and their families. For remediation to be most beneficial, one must understand and utilize areas of skill, and leverage those to positively impact deficits.

Developmental differences in brain activation of 9- to 15-year-old children were examined during an auditory rhyme decision task to spoken words using functional magnetic resonance imaging (fMRI). As a group, children showed activation in left superior/middle temporal gyri (BA 22, 21), right middle temporal gyrus (BA 21), dorsal (BA 45, pars opercularis) and ventral (BA 46, pars triangularis) aspects of left inferior frontal gyrus, and left fusiform gyrus (BA 37). There was a developmental increase in activation in left middle temporal gyrus (BA 22) across all lexical conditions, suggesting that automatic semantic processing increases with age regardless of task demands. Activation in left dorsal inferior frontal gyrus also showed developmental increases for the conflicting (e.g. PINT-MINT) compared to the non-conflicting (e.g. PRESS-LIST) non-rhyming conditions, indicating that this area becomes increasingly involved in strategic phonological processing in the face of conflicting orthographic and phonological representations. Left inferior temporal/fusiform gyrus (BA 37) activation was also greater for the conflicting (e.g. PINT-MINT) condition, and a developmental increase was found in the positive relationship between individuals' reaction time and activation in left lingual/fusiform gyrus (BA 18) in this condition, indicating an age-related increase in the association between longer reaction times and greater visual-orthographic processing in this conflicting condition. These results suggest that orthographic processing is automatically engaged by children in a task that does not require access to orthographic information for correct performance, especially when orthographic and phonological representations conflict, and especially for longer response latencies in older children.

The objective of this study was to examine the neural correlates of phonological inconsistency (relationship of spelling to sound) and orthographic inconsistency (relationship of sound to spelling) in visual word processing using functional magnetic resonance imaging (fMRI). Children (9- to 15-year-old) performed a rhyming and spelling task in which two words were presented sequentially in the visual modality. Consistent with previous studies in adults, higher phonological inconsistency was associated with greater activation in several regions including left inferior frontal gyrus and medial frontal gyrus/anterior cingulate cortex. We additionally demonstrated an effect of orthographic inconsistency in these same areas, suggesting that these regions are involved in the integration of orthographic and phonological information and, with respect to the medial frontal/anterior cingulate, greater demands on executive function. Higher phonological and orthographic consistency was associated with greater activation in precuneus/posterior cingulate cortex, the putative steady state system active during resting, suggesting lower demands on cognitive resources for consistent items. Both consistency effects were larger for the rhyming compared with the spelling task suggesting greater demands of integrating spelling and sound in the former task. Finally, accuracy on the rhyming task was negatively correlated with the consistency effect in left fusiform gyrus. In particular, this region showed insensitivity to consistency in low performers, sensitivity to inconsistency (higher activity) in moderate performers, and sensitivity to inconsistency (high activation) and to consistency (deactivation). In general, these results show that the influence of spelling-sound (and sound-spelling) correspondences on processing in fusiform gyrus develops as a function of skill.

Previous studies have shown that the strength of connectivity between regions can vary depending upon the cognitive demands of a task. In this study, the location of task-dependent connectivity from the primary visual cortex (V1) was examined in 43 children (ages 9-15) performing visual tasks; connectivity maxima were identified for a visual task requiring a linguistic (orthographic) judgment. Age, sex, and verbal IQ interacted to affect maxima location. Increases in age and verbal IQ produced similar shifts in maxima location; in girls, connectivity maxima shifted primarily laterally within the left temporal lobe, whereas the shift was primarily posterior within occipital cortex among boys. A composite map across all subjects shows an expansion in the area of connectivity with age. Results show that the location of visual/linguistic connectivity varies systematically during development, suggesting that both sex differences and developmental changes in V1 connectivity are related to linguistic function.

In academic courses in which one task for the students is to understand empirical methodology and the nature of scientific inquiry, the ability of students to create and implement their own experiments allows them to take intellectual ownership of, and greatly facilitates, the learning process. The Psychology Experiment Authoring Kit (PEAK) is a novel spreadsheet-based interface allowing students and researchers with rudimentary spreadsheet skills to create cognitive and cognitive neuroscience experiments in minutes. Students fill in a spreadsheet listing of independent variables and stimuli, insert columns that represent experimental objects such as slides (presenting text, pictures, and sounds) and feedback displays to create complete experiments, all within a single spreadsheet. The application then executes experiments with centisecond precision. Formal usability testing was done in two stages: (1) detailed coding of 10 individual subjects in one-on-one experimenter/subject videotaped sessions and (2) classroom testing of 64 undergraduates. In both individual and classroom testing, the students learned to effectively use PEAK within 2 h, and were able to create a lexical decision experiment in under 10 min. Findings from the individual testing in Stage 1 resulted in significant changes to documentation and training materials and identification of bugs to be corrected. Stage 2 testing identified additional bugs to be corrected and new features to be considered to facilitate student understanding of the experiment model. Such testing will improve the approach with each semester. The students were typically able to create their own projects in 2 h.
