Under these instruments for both domesticated (i.e. crops varieties, landraces, neglected and underutilized domesticated species) and wild PGRFA, Contracting Parties should develop their own capacity for their conservation and sustainable use in supporting food security and poverty alleviation under the Millennium Development Goals. The scope of this paper is to assess the cyto-and genotoxic effects of sulphate on Allium cepa mi... more The scope of this paper is to assess the cyto-and genotoxic effects of sulphate on Allium cepa mitosis for root meristem. Three different concentrations of sodium sulphate (i.e. 0,1%; 1% and 5%) have been used, in which the onion bulbs were immersed for different periods of time such as 6, 24 and 72 hours. In the end of the experiment the harvested root tips were prepared according to Feulgen's squash technique and using Schiff reagent. The cytotoxic effects of sulphate were investigated by calculating the mitotic index and also through the analysis of chromosomes alterations during the mitosis. The phase ratio of cells undergoing mitosis in all the phases is estimated for all variants. For microscopy investigations it was used a Novex Holland B microscope with digital camera included. The cytological analysis realized on Allium cepa revealed a strong decrease in the mitotic index due to sulphate treatments which is more intense with the time of exposure. Moreover this phenomenon is associated with the appearance of different chromosomal complement alterations including the appearance of highly condensed chromatin. The mitotic index and genotoxic observations over the chromosomes can also be correlated with phase ratio of cells undergoing mitosis.

The following determinations were accomplished on a portion of the visceral mass for its physic-chemical characterization: the determination of loss by drying, the determination of calcinations residue, the determination of total protein through the Kjeldahl method. The other portion of the visceral mass was used to obtain proteinaceous extracts 1: 3 and 1: 5 on which the following determinations were made: the determination of loss by drying, the determination of total protein through the Lowry method, the determination of pH, the Triticum test for the evaluation of the cytotoxic and cytostatic properties of the proteinaceous extract obtained from the visceral mass of mollusks. On farm conservation of crop landraces or old traditional crops is essential for further supporti... more On farm conservation of crop landraces or old traditional crops is essential for further supporting food security and new policies in relation with biodiversity conservation and climate change responses such as adaptation and mitigation. In order to improve the genetic resources knowledge regarding crop landraces’ germplasm diversity a new methodology for assessing their status of conservation thorough a landscape approach by also taking into account the climate change vulnerability of certain ecosystems compared to others and specific environment requirements is proposed. Developing new policies in applying dynamic action plans in agriculture, according to environmental factors, climate change impact and tolerance degree of crop landraces, may originate from these types of integrated studies. Based on the above mentioned rationale this proposed methodology will be further used for developing a Red List for crops in Romania.

A micropropagation protocol for spathiphyllum is described in this article. The original material... more A micropropagation protocol for spathiphyllum is described in this article. The original material started from meristem culture and a dynamic of the second to the fourth multiplication stages was analyzed. An exponential growth with a correlation index of R2 = 0.9916 was observed. The multiplication index was positively influenced by the incorporation into the culture medium of BAP (1 mg/D and IAA (0.1 mg/D which gave 26.6 new shoots/explant. A culture medium supplemented only with auxin did not induce callus formation and when only cytokinin was used callusogenesis was positively influenced and rooting processes were not observed.

One problem we face in recent years is the raw material derived from this species, which is quit few compared with the requirements. A cause for this situation is the germinative faculty, which Angelica archangelica L. seeds squickly lose. This is in contradiction with the fact that when establishing any crop by direct sowing, the quality of material used for seeding has a high importance. So, we consider important at this stage any study action taken for identifying problems that generate this phenomenon. This article examines the influence which umbel type and its period of occurrence, of which we harvest the seeds, has on the process of germination. A data sheet model this paper is proposing that will be required for developing a future Red List... more A data sheet model this paper is proposing that will be required for developing a future Red List for crop landraces in Romania. Such a Red List is not yet published in our country and the genetic erosion for crops is increasing especially because of the pressure of commercial crops entering the market-place and also because of the climate change and desertification as major threats. As a consequence for safeguarding food and feed it is compulsory to preserving genetic resources and a special attention should be devoted to on farm conservation. Developing a red list for crop landraces in Romania will support further on farm conservation of such crops and will more emphasize the role of gene banks in our country. Furthermore such a red list will ground the development of a new agriculture vision and policy regarding the implementation of appropriate incentive measures for supporting on farm conservation of crop landraces in specific area in Romania such is the protected area system.

The cytogenetic effects were studied by the calculation of the mitotic index, by the study of the interphase and chromosomal aberrations on the mitotic cells. We used MnSO4 and Pb (NO3) 2 solutions with different concentrations: 0.0003, 0.003, and 0.03%. The Allium bulbs were preliminary imbued in water, and then they were treated for 12, 24 and respectively 72 hours in these solutions.
