Method: Sorafenib-resistant cells were generated by treating naïve cells (HepG2215 and Hep3B) with sorafenib. Different cancer cell lines from databases were analyzed through the ONCOMINE web server. BIOSTORM–LIHC patient tissues (46 nonresponders and 21 responders to sorafenib) were used to compare YAP mRNA levels. The HepG2215_R-derived xenograft in SCID mice was used as an in vivo model. HCC tissues from a patient with sorafenib failure were used to examine differences in YAP and IGF-R signaling. Results: Positive associations exist among the levels of YAP, IGF-1R, and EMT markers in HCC tissues and the levels of these proteins increased with sorafenib failure, with a trend of tumor-margin distribution in vivo. Blocking YAP downregulated IGF-1R signaling-related proteins, while IGF-1/2 treatment enhanced the nuclear translocation of YAP in HCC cells through PI3K-mTOR regulation. The combinatio...

We assessed attitudes toward at-home HPV self-collection compared with clinic-based Pap testing in this higher-risk population. Participants were low-income women in North Carolina overdue for cervical cancer screening. Women self-collected samples at home, returned samples by mail for HPV testing, and completed phone questionnaires about at-home HPV self-collection. Participants were referred to clinic-based Pap testing and invited to complete a second questionnaire about Pap testing. A cross-sectional questionnaire compared attitudes, experiences, and preferences for self-collection versus Pap testing and assessed predictors of preference for HPV self-collection. The prevalence and regulation of p38 mitogen activated protein kinase (MAPK) expression in human ... more The prevalence and regulation of p38 mitogen activated protein kinase (MAPK) expression in human lymphomas have not been extensively studied. In order to elucidate the role of p38 MAPK in lymphomagenesis, we examined the expression of native and phosphorylated p38 (p-p38) MAPK in cell lines derived from human hematopoietic neoplasms including B cell lymphoma-derived cell lines using Western blot analysis. The p-p38 MAPK protein was also analyzed in 30 B cell non-Hodgkin lymphoma (NHL) tissue biopsies by immunohistochemistry. Our results show that the expression of p38 MAPK was upregulated in most of the cell lines as compared with peripheral blood lymphocytes, while the expression of p-p38 MAPK was more variable. A subset of B cell NHL biopsies showed increased expression of p-p38 MAPK relative to reactive germinal center cells. Interleukin-4 (IL-4) induced a dose-dependent increase in the expression of p-p38 MAPK (1.6-to 2.8-fold) in cell lines derived from activated B cell-like diffuse large B cell lymphoma (DLBCL) but not those from germinal center-like DLBCL. No change was seen in native p38 MAPK. The in vitro kinase activity of p38 MAPK, however, was induced (1.6to 3.2-fold) in all five cell lines by IL-4. Quantitative fluorescent RT-PCR demonstrated that all four isoforms of p38 MAPK gene were expressed in the lymphoma cell lines, with p38γ and p38β isoforms being predominant. IL-4 stimulation increased the expression of β, γ, and δ isoforms but not α isoform in two cell lines. In conclusion, there is constitutive expression and activation of p38 MAPK in a large number of B-lymphoma-derived cell lines and primary lymphoma tissues, supportive of its role in lymphomagenesis. The differential IL-4 regulation of p38 MAPK expression in cell lines derived from DLBCL may relate to the cellular origin of these neoplasms.

A nonperturbing probe that utilizes a focused ion beam has been developed to measure the position... more A nonperturbing probe that utilizes a focused ion beam has been developed to measure the position and profile of a tightly bunched charged particle beam (CPB). The ions are injected across the path of the CPB and are deflected by the electromagnetic field of the CPB. Deflected ions are detected by a gated microchannel plate detector and video camera. From the magnitude of the deflections, one can determine the position, radial profile, and in some cases, the axial length of the CPB. Ion parameters can be adjusted to tailor the probe to a wide range of CPB parameters, from submicron, ps bunches to millimeter-sized, quasicontinuous beams. We present the theory of operation of the probe, provide a number of numerical examples to illustrate its capabilities, and describe a proof-of-principle version of the probe that we built and successfully tested at SLAC. Extensive experimental measurements were carried out to test the accuracy of the parametric helix... more Extensive experimental measurements were carried out to test the accuracy of the parametric helix traveling-wave tube (TWT) code, CHRISTINE [1]. The model is one-dimensional, with beam electrons represented as rigid disks. Multifrequency interactions are supported and the RF circuit can be optionally represented with cold-test data, a sheath helix model, or a recently implemented tape helix model [2]. Simulations using the tape helix model are shown to be in good agreement with experimental measurements of an L-band TWT over a broad (250-MHz) frequency range. In the intermediate and saturated power regimes, the modeled and measured TWT gain versus frequency agree to better than 0.4 dB, with deviations explained by strong reflections at the output window that are not accounted for in the code. Single-tone experimental and simulated drive curves agree to better than 1 dB in the small-and large-signal regimes; relative phase shift simulations agree to within experimental measurement accuracy in the small-signal regime and to within 75% in the large-signal regime. Two-tone experimental and modeled data exhibit similarly good agreement, with CHRISTINE accurately predicting the effect of frequency-dependent gain variations on the TWT output response and third-and fifth-order intermodulation products. INTRODUCTION T ELECOMMUNICATIONS, radar, and electronic warfare applications of traveling-wave tube (TWT) amplifiers often require the simultaneous amplification of multiple-frequency signals. However, nonlinearities in the amplifier can lead to the creation of unwanted interaction products, such as amplitude and phase cross modulation and intermodulation. These interaction products reduce the power available to the driven signals and can potentially distort driven signal amplitudes and phases, leading to a degradation in performance, e.g., increased bit-error rates in communications systems. As operating bandwidths and channel densities within bands increase, the problem is exacerbated.
