isolation: Leaves and stem portions of different medicinal plants [Ocimum tenuiflorum L., Ocimum basilicum L., Mentha spicata L.., Vitex negundo L., Plectranthus amboinicus (Lour.) Spreng., Anisomeles malabarica (L.) The last surface-sterilized water was plated on media for the growth of microbes. The effectiveness of surface sterilization was detected by the lack of microbial growth on the inoculated plate. Surface sterilized leaves and stems were cutinto small pieces (1 cm x1 cm) placed on potato dextrose agar (PDA) media supplemented with a pinch of streptomycin and incubated at 25°C. The observation was made from the second day up to 3-4 weeks for fungal colonization’®. The pure culture of endophytic fungi grown from internal tissues was tested for purity before being transferred to new PDA slants and preserved at 4°C for further experiments.

Figure 3: Quantitative screening of L-Asparaginase enzyme activity by promising endophytic fungi cultures in Modified Czapek’s Dox broth L-Asparaginase activity: Among the 40 endophytic fungi tested for L-Asparaginase, 52.5% of isolates revealed positive enzyme activity. L-Asparaginase is a therapeutic enzyme used as a chemotherapeutic drug to cure acute lymphoblastic leukemia!’. Nine fungal endophytes isolated from different medicinal plants present in the high mountains of Uttarakhand, India indicated positive L- Asparaginase activity*°. El-Said et al'® tested six Aspergillus niger isolates for L-Asparaginase production. All the six tested isolates were capable of synthesizing L-A sparaginase with moderate activity. 89 endophytic fungi were isolated from six different Thymus sp. and further screened for L- Jenila and Gnanadoss?? studied the maximum L- Asparaginase activity of 5.74+1.2 U/mL and 6.07+0.8 U/mL for the cultures LCJ273 and LCJ294 on the 6" day in Czapek’s Dox medium. Fusarium tricinctum obtained from the stem segment of Tabernaemontana heyneana displayed a maximum of 3.423+0.05 U/mL L-Asparaginase activity°.
