The jumbo or Humboldt squid, Dosidicus gigas, is an important fisheries resource and a significant participant in regional ecologies as both predator and prey. It is the largest species in the oceanic squid family Ommastrephidae and has the largest known potential fecundity of any cephalopod, yet little is understood about its reproductive biology. We report the first discovery of a naturally deposited egg mass of Dosidicus gigas, as well as the first spawning of eggs in captivity. The egg mass was found in warm water (25 -278C) at a depth of 16 m and was far larger than the egg masses of any squid species previously reported. Eggs were embedded in a watery, gelatinous matrix and were individually surrounded by a unique envelope external to the chorion. This envelope was present in both wild and captive-spawned egg masses, but it was not present in artificially fertilized eggs. The wild egg mass appeared to be resistant to microbial infection, unlike the incomplete and damaged egg masses spawned in captivity, suggesting that the intact egg mass protects the eggs within. Chorion expansion was also more extensive in the wild egg mass. Hatchling behaviours included proboscis extension, chromatophore activity, and a range of swimming speeds that may allow them to exercise some control over their distribution in the wild.

Artificial insemination of the Argentine shortfin squid Illex argentinus was carried out on board, and the hatchlings were successfully obtained. Applying oviducal gland jelly to inseminated egg al lowed the embryo to expand chorion and develop perivitelline space, but applying nidamental gland jel ly and applying no jelly was not successful. The eggs which developed perivitelline space only continued embryonic development normally and proceeded onto hatching. The embryonic development was ob served in vitro throughout the experiment. The hatchlings survived for about one week in vitro without feeding.

Oocyte development, fecundity and spawning strategy of large sized jumbo squid Dosidicus gigas (Oegopsida: Ommastrephinae

Ovaries of 39 large jumbo squid Dosidicus gigas (415– 875 mm mantle length (ML)) of different stages of maturity from the Gulf of California and Nicaragua were analysed. Oocyte development is asynchronous during female ontogenesis, with predominance (.85 –90%) of small protoplasmic oocytes 0.1 –0.2 mm in diameter for all stages of female maturity. Potential fecundity (PF) is determined at late immature stage II with average values around 18– 21 million oocytes. The maximum PF estimated, 32 million oocytes, is so far the largest recorded for any cephalopod. PF is closely related to female size. Nicaraguan females, of smaller size (415– 720 mm ML), show lower fecundity (5– 15 million oocytes). Eggs were analysed in oviducts of mature females (540– 875 mm ML), 7 from the Gulf of California and three females from Nicaragua. Eggs measured 0.9 Â 1.1 mm in diameter and weighed 0.45 mg on average. The oviducts of the largest female (875 mm ML) held 1.2 million eggs, representing 4.6% of PF and no more than 14% of all vitelline oocyte stock of that squid. Variability in oviduct filling is very high and unrelated to ovary weight. However, a strong correlation was found between oviduct fullness and nidamental glands development. These observations suggest that spawning is extended and intermittent. A female spawns no less than half of the initial PF, and the minimal number of spawning activity events (egg batches) is roughly estimated at 8–12. During this terminal spawning stage females continue actively feeding and grow between egg-mass laying periods.

Early life stages of cephalopods are somewhat complex due to the life history strategy or species specificity of generalized ontogenetic patterns and processes. This work aimed to determine the time length of embryonic development at different temperatures, and if the egg size is a determinant of hatchling size in Sepiola atlantica d 0 Orbigny, 1839-1842. Successful hatching occurred in 98.5-100% of the eggs for each female. As seen in other coleoid cephalopods, temperature determines the amount of time for embryonic development in S. atlantica, and the obtained data were very similar to other coleoid cephalopods. Developmental times for temperatures at 13 ± 0.4°C, 18 ± 0.3°C and 16.4 ± 1.1°C were 61.8 ± 3.8, 22.6 ± 1.7 and 40.1 ± 4.8 days. The duration of embryonic development and hatchling mantle length was not strictly related. The egg volume was positively related to hatchling mantle length. Our results provide new records on the duration of embryogenesis and other information on reproductive patterns in this species. Some hatching and post-hatching behaviour are shown and discussed.

The effects of egg position, egg mass size, substrate and biofouling on embryo mortality in the squid Sepioteuthis australis

Using a combination of laboratory and field investigations, this study examined embryo mortality in the southern calamary Sepioteuthis australis as a function of egg mass size, the substrate upon which the mass is attached, the position of the embryo within the mass, and the degree of biofouling. Egg mass size ranged from 2 to 1,241 egg strands, however most masses consisted of 200-299 strands. Small egg masses (<300 strands) were generally attached to soft-sediment vegetation (Amphibolis antarctica, Heterozostera tasmanica, Caulerpa sp.), whereas larger masses (>300 strands) were either securely attached to robust macroalgae holdfasts (Ecklonia sp., Marcocystis pyrifera, Sargassum sp.) or unattached. Rates of embryo mortality were highly variable ranging from 2 to 25%. Both laboratory and field results indicated a positive relationship between egg mass size and embryo mortality. Larger, unattached egg masses contained twice as many dead embryos than those securely attached to a substrate. Mortality rates were significantly affected by the embryos' relative position within the mass and were highest in embryos located near the attachment point of the egg strand, within the interior of the mass, and in close contact with the substrate. This was attributed to the inability of the embryos to respire adequately and eliminate metabolic wastes. Biofouling did not strongly influence embryo mortality, but colonisation occurred in areas conducive to growth, photosynthesis, and respiration indicating 'healthy' regions within the mass.

Embryonic life of the loliginid squid Loligo vulgaris: comparison between statoliths of Atlantic and Mediterranean populations

Egg masses of the loliginid squid Loligo vulgaris Lamarck, 1798 are attached to hard substratum or branched sessile organisms on the sea bottom. Embryonic development lasts from a few weeks to a few months, depending on the environmental water temperature. Because embryonic statolith growth of L. vulgaris is very sensitive to temperature under laboratory conditions, we analyzed the possibilities of determining past events in the squid's early life from analysis of the embryonic area of statoliths of wild squid populations. The relationship between egg-incubation temperature and daily growth of embryonic statoliths under laboratory conditions was determined by tetracycline markings at 10 incubation temperatures ranging from 12 to 24.7°C. In addition, the mean width of embryonic increments in statolith collections of wild L. vulgaris from the Eastern Atlantic (Saharan Bank and NW Iberian Peninsula) and the Mediterranean Sea (Central and Eastern) was calculated. The temperature inferred from the embryonic increment widths of the statoliths of wild squid indicates that embryonic development of L. vulgaris in the regions sampled is likely to occur at temperatures ranging from 12 to 17°C. Mediterranean squid have wider embryonic increments than Atlantic squid, reflecting the slightly higher water temperatures in the Mediterranean Sea during the development of the egg masses. Eggs of L. vulgaris spawned off the NW Iberian Peninsula were estimated, on average, to remain at sea for 47 d, 1 wk longer than Mediterranean eggs (nearly 1 mo longer when comparing minimum and maximum ranges). A longer incubation time for egg masses attached to the sea bottom increases mortality risks. Conversely, slow development at a lower temperature can improve yolk conversion, producing larger hatchlings, and increased hatching competence is expected from such squid. Therefore, a compromise between longer-versus-shorter incubation time and related characteristics does exist.

Egg masses of oceanic squid accidently collected in the wild have been observed only from a few spawning events in aquaria, and as a consequence, the study of their embryos and hatchlings is very limited. Here, we used in vitro fertilization techniques to understand the abiotic factors that influenced egg development and hatchling performance of the ommastrephid squid Illex coindetii from the Mediterranean Sea. Egg and hatchling sizes of these ommastrephids are close to the minimum for cephalopod species, and therefore, short embryonic developmental periods were expected under a Mediterranean temperature regime. Low incubation densities and the use of antibiotics in the incubation medium resulted in relatively high survival rates to hatchling. Hatching time was inversely proportional to incubation temperature and ranged from 5 to 11 days at 21°C and 13°C, respectively. The addition of oviducal jelly to the eggs immediately after fertilization, followed by another addition at the beginning of organogenesis, resulted in better chorionic expansion, with an increase in egg diameter, delayed hatching and higher hatchling weight, which suggests a better use of yolk reserves. Larger hatchlings in length and weight also tended to be produced at low temperature, probably also due to better yolk absorption. As this species spawns all year round, size differences between summer and winter hatchlings may be suspected. In vitro fertilization techniques used in the laboratory proved to be useful in shedding some light on the early life of this oceanic squid. Further research is needed to elaborate on tests to predict gamete quality and to develop methods to avoid premature hatching.

Useful Techniques for Artificial Fertilization of the Ommastrephid Squid Illex argentinus

This study examined the rate of fertilization in the Argentine shortfin squid Illex argentinus after artificial insemination using sperm from different sources, as well as the effect of cold-storage on the fertilization capacity of gametes from mature squids, and the impact of mechanical agitation on embryonic development. In addition, the process of chorionic expansion during the course of embryonic development was also examined. High fertilization rates were obtained from spermatophores (76-95%) and spermatozoa from the vas deferens (73-87%), but no fertilization was observed using spermatozoa from testes. Female and male gametes stored at 0°C had a high fertilization capacity (>72%) within 24 h. Embryos (stages 4 to 17; 3 to 61 h at 20 o C) subjected to mechanical agitation at a critical period (stages 5 to 12; 5 to 20 h at 20 o C) exhibited high mortality and abnormal development. In the course of embryonic development, the chorion expanded four times: once at fertilization and then at developmental stages 15-16, 17-21, and finally at stages 25-27. The growth rate of the third expansion corresponded with an increase in embryonic body length. To improve the fertilization rate and increase the production of healthy paralarvae in on-board experiments, factors such as the origin of spermatozoa, gamete storage methods, and the handling of fertilized eggs during the critical period should be carefully controlled.

Observations on embryos and embryonic development from an egg mass of the jumbo squid Dosidicus gigas spawned under captive conditions

Egg masses were spawned by a jumbo squid Dosidicus gigas (mantle length 37.5 cm) held in a tank (500 L) on board the R/V Kaiyo Maru during a joint Japan-Peru cruise in Peruvian waters during December 2011-February 2012. Part of an egg mass was collected and incubated in an aquarium (10 L) maintained at 20 °C. The eggs had a unique jelly envelope surrounding the chorion. The diameter of the jelly envelope was more than twice the diameter of chorion. It remained clearly visible until the embryos reached developmental stage 18. Most of the eggs were fertilized and hatched (Stage 30) 6.5 days after spawning at 20 °C.

    Early Mode of Life and Hatchling Size in Cephalopod Molluscs: Influence on the Species Distributional Ranges

    Cephalopods (nautiluses, cuttlefishes, squids and octopuses) exhibit direct development and display two major developmental modes: planktonic and benthic. Planktonic hatchlings are small and go through some degree of morphological changes during the planktonic phase, which can last from days to months, with ocean currents enhancing their dispersal capacity. Benthic hatchlings are usually large, miniature-like adults and have comparatively reduced dispersal potential. We examined the relationship between early developmental mode, hatchling size and species latitudinal distribution range of 110 species hatched in the laboratory, which represent 13% of the total number of live cephalopod species described to date. Results showed that species with planktonic hatchlings reach broader distributional ranges in comparison with species with benthic hatchlings. In addition, squids and octopods follow an inverse relationship between hatchling size and species latitudinal distribution. In both groups, species with smaller hatchlings have broader latitudinal distribution ranges. Thus, squid and octopod species with larger hatchlings have latitudinal distributions of comparatively minor extension. This pattern also emerges when all species are grouped by genus (n = 41), but was not detected for cuttlefishes, a group composed mainly of species with large and benthic hatchlings. However, when hatchling size was compared to adult size, it was observed that the smaller the hatchlings, the broader the latitudinal distributional range of the species for cuttlefishes, squids and octopuses. This was also valid for all cephalopod species with benthic hatchlings pooled together. Hatchling size and associated developmental mode and dispersal potential seem to be main influential factors in determining the distributional range of cephalopods.

    Predatory flying squids are detritivores during their early planktonic life

    Cephalopods are primarily active predators throughout life. Flying squids (family Ommastrephidae) represents the most widely distributed and ecologically important family of cephalopods. While the diets of adult flying squids have been extensively studied, the first feeding diet of early paralarvae remains a mystery. The morphology of this ontogenetic stage notably differs from other cephalopod paralarvae, suggesting a different feeding strategy. Here, a combination of Laser Capture Microdissection (LCM) and DNA metabarcoding of wild-collected paralarvae gut contents for eukaryotic 18S v9 and prokaryotic 16S rRNA was applied, covering almost every life domain. The gut contents were mainly composed by fungus, plants, algae and animals of marine and terrestrial origin, as well as eukaryotic and prokaryotic microorganisms commonly found in fecal pellets and particulate organic matter.     Multiplicity of sperm receptacles for securing the offspring genetic variability in a pelagic squid

    Female eumetazoans often develop sperm storage organs (SSOs). Although the processes of sperm storage may in uence sperm competition and cryptic female choice in polyandrous species, the signi cance of developing multiple SSOs is not well understood. In contrast to coastal squids (which develop no more than two SSOs), the female Japanese common squid Todarodes paci cus, a more oceanic pelagic species, develops more than 20 SSOs, which take the form of specialized pockets, called seminal receptacles (SRs), on the buccal membrane. We investigated the sperm storage pattern of SRs by paternity analysis of hatchlings obtained after arti cial insemination using sperm retrieved from 6 arbitrarily selected SRs. The results showed that females were capable of storing sperm contributed by 9 to 23 males, indicating that females are broadly promiscuous. In the pattern of sperm storage, the number of males and proportion of their sperm present in the SRs varied widely among SRs, and sperm storage was biased towards particular males at the individual SR level. However, when calculated as a proportion of all the SRs within a female, the number of sires increased and the paternity bias towards any particular male weakened. These results suggest that one function of having multiple SRs in T. paci cus may be to ensure genetic diversity of the offspring.
