A series of experiments were conducted in Plant Two local wheat varieties cultivars named Dor29 a... more A series of experiments were conducted in Plant Two local wheat varieties cultivars named Dor29 and Fateh were evaluated to determine the impact of 0, 10, 20 and 30 g.lP-1 P sorbitol for callus induction and somatic embryos regeneration. Mature seeds of the tested cultivars were selected as explants for callus induction and regeneration on MS medium with Gambrog vitamins containing 30 g.lP-1 P sucrose, 5 g.lP-1 P agar, 2 mg.lP-1 P BA and 1 mg.lP-1 P 2,4-D. Results were showed that sorbitol at 20 g.lP-1 P had a significant effect on callus fresh weight; mean globular somatic embryos number with no significant difference between the two cultivars under investigation. The obtained results from our study suggested that the optimal callus induction medium was MS + Gambrog vitamins + 30 g.lP-1 P sucrose + 20 g.lP-1 P sorbitol + 2 mg.lP-1 P BA + 1 mgP-P 2,4-D + 5 g.lP-1 P agar ,while maximum number of somatic embryos formed at the same previous medium with the amition of BA and 2,4-D.

The study was conducted to determine the appropriate concentration of coconut water (CW) that enh... more The study was conducted to determine the appropriate concentration of coconut water (CW) that enhance an efficient and cost effective protocol for rice micropropagation through callus induction and somatic embryogenesis.Seeds of a local Iraqi rice (Oryza sativa L.) cultivar Amber33, were used as explant which inoculated on basal medium consists of MS inorganic salts, Gamborg vitamins and supplemented with the auxin 2,4-Dichlorophenoxyacetic acid (2,4-D) and the cytokinin Benzyl adenine (BA). In order to evaluate the role of CW, three concentrations (10, 20 and 30 g.l-1) were added to MS inorganic salts after the exclusion of growth regulators (2.4-D and BA) and Gamborg vitamins. Results showed that adding CW especially at concentration of 30g.l-1 have minimized the callus induction period, enhanced callus fresh weight and numbers and germination percentage of somatic embryos when compared with the media enriched with growth regulators only.

Baghdad. Seeds were soaked with three concentrations of SA (0.1,0.3 and 0.5 mM) in addition to control treatment with distilled water, while PEG8000 were used at concentrations of 0% (distilled water) 1%, 2%, 3%, 6%, 9% and 12%. Callus induction and plant regeneration through somatic embryogenesis in Phalaenopsis Richard Sha... more Callus induction and plant regeneration through somatic embryogenesis in Phalaenopsis Richard Shaffer 'Santa Cruz' were examined. Protocorm-like body (PLB) segments formed calli in Vacin and Went medium with sucrose. The optimal concentration of sucrose was 40 g • l-1. Medium containing 200 ml • l-1 coconut water together with 40 g • l-1 sucrose was effective for callus induction. Gellan gum was suitable than agar as a gelling agent for callus induction. The calli easily formed PLBs after being transferred to a medium without sucrose. Histological observation suggested that the PLBs were somatic embryos. No variation was observed in the flowering plants regenerated through somatic embryogenesis.

Lead as lead nitrate (Pb(NO3)2) was added alone to the pots at concentrations of 0 (full strength Hoagland nutrient solution), 0.25, 0.5 and 0.75 mM or in interaction with EDTA at concentrations of 0, 5 and 10 mM. The soaking of seeds with H O was carried out at 1.5 mmol concentration in addition to 2 2 distilled water (control) for 6 hours and the seeds were sown in pots contain on mixture river sand and peat. The seedling was exposed after two weeks from date of germination for induced salt stress with NaCl at different concentrations (50, 100, 150 mmol) for 8 weeks. The salt stress led to significant decrease in shoot height, leaf area, fresh and dry weight, relative water content and significant increase activity of catalase (CAT) and superoxide dismutase (SOD) at 150 mmol concentration, while increase was observed in activity of ascorbate peroxidase (APX) at 50 mmol concentration. The salt stress caused significant decrease in concentration of ascorbic acid but with no difference in concentration of glutathione and carotene. Salt stress caused significant increase in concentration of proline and malondialdehyde (MDA) content and increase of electrolyte leakage. The pretreatment of seeds with H O led to improvement in some of growth parameters (leaf area and fresh weight) and 2 2 improvement in enzymatic antioxidants (CAT, SOD, APX) and also in enhancement of non-enzymatic antioxidants(carotene, glutathione, ascorbic acid) levels and reduction in MDA content and electrolyte leakage.

The effect of seawater salinity on callus growth, sodium and potassium concentrations, proline ac... more The effect of seawater salinity on callus growth, sodium and potassium concentrations, proline accumulation and some antioxidant enzymes ( ascorbate peroxidase, catalase and superoxide dismutase) activity in the callus of tomato (Lycopersicon esculentum Mill.) cv. Super Marmmande were investigated. Seeds of tomato were used as explant and cultured using Pyrex test tubes (150x20 mm) on MS medium with Gamborg vitamins, sucrose, 2,4-D, BA and soldifies with agar, for six weeks, to induce the callus formation and to get a considerable amount of callus to be used in the upcoming experiment. Seawater was diluted with distilled water and added to the medium at three concentrations (20%, 30% and 40%), 100 mg of the induced callus were cultured on the same previous medium for another six weeks to evaluate the effects of seawater salinity on growth and some biochemical parameters. Results indicated that high seawater concentration (40%) caused a significant decrease in callus fresh weight and potassium ion concentration, while a significant increase noticed in sodium ion concentration, proline accumulation, and the activity of antioxidant enzymes in the callus.
