In this paper, the optimization of laser-MAG hybrid welding parameters of 10CrNi3MoV ship steel w... more In this paper, the optimization of laser-MAG hybrid welding parameters of 10CrNi3MoV ship steel was developed. Using the Box-Behnken Design (BBD) model in Response Surface Methodology (RSM) and taking laser power, welding speed and welding current as response factors, the design matrix was completed and verified by experiment. The regression model associated with welding parameters was established by measuring the response indices, such as penetration, tensile strength and impact absorption energy. Through the model check, it was found that the accuracy of penetration and tensile strength of the model was high, and the optimized parameters were as follows: laser power (P) = 3700 W, welding speed (V) = 0.8 m/min, wire feeding speed (Vs) = 7 m/min. On the premise of meeting mechanical performance inspection standards, the maximum penetration was 8 mm.

Cancer stemness represents a major source of development and progression of colorectal cancer (CR... more Cancer stemness represents a major source of development and progression of colorectal cancer (CRC). c-Met critically contributes to CRC stemness, but how c-Met is activated in CRC remains elusive. We previously identified the lipolytic factor ABHD5 as an important tumour suppressor gene in CRC. Here, we show that loss of ABHD5 promotes c-Met activation to sustain CRC stemness in a non-canonical manner. Mechanistically, we demonstrate that ABHD5 interacts in the cytoplasm with the core subunit of the SET1A methyltransferase complex, DPY30, thereby inhibiting the nuclear translocation of DPY30 and activity of SET1A. In the absence of ABHD5, DPY30 translocates to the nucleus and supports SET1A-mediated methylation of YAP and histone H3, which sequesters YAP in the nucleus and increases chromatin accessibility to synergistically promote YAP-induced transcription of c-Met, thus promoting the stemness of CRC cells. During the process of reeling and unreeling, pipelines are subjected to nominal strain of 1-4%. In this study, various pipeline geometries and crack dimensions were calculated using a finite element analysis and analytical solution under strain-controlled boundary conditions. A modified reference stress approach was applied under strain-controlled boundary conditions using a derived implicit analytical solution of the nominal stress. The limit load solution was modified through a γ factor based on the finite element analysis under strain-controlled boundary conditions. A regression analysis of γ was conducted and an empirical formula was determined. The modified strain-controlled reference approach is in contrast to other ECA methods and proved to be closer to the finite element analysis results.

Nup358 binds and activates the auto-inhibited dynein adaptor Bicaudal D2 (BicD2), which in turn recruits and activates the dynein machinery to position the nucleus. However, the molecular details of the Nup358/BicD2 interaction remain poorly understood. Here, we show that a minimal Nup358 domain activates dynein/dynactin/BicD2 for processive motility on microtubules. Using nuclear magnetic resonance (NMR) titration and chemical exchange saturation transfer (CEST), a Nup358 helix encompassing residues 2162-2184 was identified, which transitioned from random coil to an α-helix upon BicD2-binding and formed the core of the Nup358-BicD2 interface. In the offshore industry, oil and gas are commonly transported with thin-walled clad pipes. Reel-... more In the offshore industry, oil and gas are commonly transported with thin-walled clad pipes. Reel-lay is a widely used method to install clad pipes. During the installation and service process, engineering critical assessment (ECA) is usually required to guarantee safety. Most of the ECA methods pertain to surface cracks. The DNV-RP-F108 standard indicates that an embedded crack can be transformed into a surface crack in evaluation, and the result is still conservative. However, the evaluation can be over-conservative at times, meanwhile, the effect of the weld material as well as groove shape is not considered. In this work, a more direct assessment of welded clad pipes with a circumferential embedded crack is proposed based on a modified reference strain approach. A strain-controlled boundary condition is applied, and various pipe and weld geometrical shapes are considered in the analysis matrix. Two plugins based on Python script are developed to circularly create a lot of numerical models. Hundreds of regression analyses based on the linear relationship between normalized J integral and nominal strain are implemented. An analytical solution of the peak J integral of the embedded crack is proposed, and the solution is proved to be close to FEA.

Background: Ankylosing spondylitis (AS) is a chronic inflammatory rheumatic disease with a usual ... more Background: Ankylosing spondylitis (AS) is a chronic inflammatory rheumatic disease with a usual onset in the third decade of life, when persons are committed to various social roles that adults fulfil. 1 When evaluating the outcomes of clinical care, social role participation is an increasingly important outcome, especially when considering diseases with substantial limitations in physical functioning such as inflammatory rheumatic diseases. 2 The Short Form of the Social Role Participation Questionnaire (s-SRPQ) is a questionnaire which was developed to assess effect of ankylosing spondylitis (AS) on participation. 2 Objectives: This study aims to evaluate the reliability and validity of the Turkish version of the s-SRPQ in Turkish patients with AS. Methods: The Turkish version of s-SRPQ questionnaire was obtained after a translation and back translation process. The study sample included 100 AS patients (59 males, 41 females; mean age 42.00±11.00 years; range 19 to 69 years). To assess the test-retest reliability of the Turkish s-SRPQ, the questionnaire was reapplied 15 days after the first interview (interclass correlation coefficient, ICC). Cronbach's alpha (a) was used to evaluate the internal consistency. The s-SRPQ was compared with Short Form-36 survey (SF-36), Ankylosing Spondylitis Quality of Life questionnaire (ASQoL), Bath Ankylosing Spondylitis Disease Activity Index (BASDAI), Bath Ankylosing Spondylitis Functional Index (BASFI), and Satisfaction With Life Scale (SWLS) for convergent validity. The internal structure of s-SRPQ was examined by factor analysis. Results: For s-SRPQ/experienced physical difficulties; the individual item ICC ranged from 0.78 to 1.00 and Cronbach's alpha value ranged from 0.88 to 1.00. For s-SRPQ/satisfaction with role performance; the individual item ICC ranged from 0.93 to 0.98 and Cronbach's alpha value ranged from 0.96 to 0.99. KMO values was determined as 0.90 and 0.89 in the s-SRPQ/experienced physical difficulties and s-SRPQ/satisfaction with role performance, respectively. Bartlett's test of sphericity had a p<0.001. The Turkish version of s-SRPQ/experienced physical difficulties scores negatively correlated with the SWLS and SF-36 sub-parameters scores (p<0.01). There were also strong positive correlations between s-SRPQ/ experienced physical difficulties scores and BASDAI, BASFI, and ASQoL scores (p<0.01). The SRPQ/satisfaction with role performance scores positively correlated with the SWLS and SF-36 sub-parameters scores (p<0.01). There were also strong negative correlations between s-SRPQ/experienced physical difficulties scores and BASDAI, BASFI, and ASQoL scores (p<0.01). Conclusions: Turkish version of s-SRPQ has good comprehensibility, internal consistency, and validity and is an adequate and useful instrument for the assessment of participation in Turkish patients with AS.
