Present investigation entails carbonic anhydrase (CA) immobilization and its influence on transfo... more Present investigation entails carbonic anhydrase (CA) immobilization and its influence on transformation of CO 2 to calcite. CA enzyme was immobilized in alginate beads, subsequently maintained its catalytic efficiency after sequential operational cycles. The immobilized beads showed better operational stability by retaining nearly 67% of its initial activity even after six cycles. Batch scale studies with free and immobilized enzyme revealed that the entrapped CA hydrates CO 2 to bicarbonate and/or carbonate which was then made to react with Ca 2+ ions to transform into calcite. Calcite was characterized by X-ray diffraction (XRD) and scanning electron microscopy (SEM). The entrapped CA was employed for the performance evaluation with respect to several operational parameters including the influence of enzyme concentration in free and immobilized condition. It was concluded that immobilized CA in alginate beads would have the potential for CO 2 sequestration by biomimetic route.

Anthropogenic activities have substantially increased the level of greenhouse gases (GHGs) in the... more Anthropogenic activities have substantially increased the level of greenhouse gases (GHGs) in the atmosphere and are contributing significantly to the global warming. Carbon dioxide (CO2 ) is one of the major GHGs which plays a key role in the climate change. Various approaches and methodologies are under investigation to address CO2 capture and sequestration worldwide. Carbonic anhydrase (CA) mediated CO2 sequestration is one of the promising options. Therefore, the present review elaborates recent developments in CA, its immobilization and bioreactor methodologies towards CO2 sequestration using the CA enzyme. The promises and challenges associated with the efficient utilization of CA for CO2 sequestration and scale up from flask to lab-scale bioreactor are critically discussed. Finally, the current review also recommends the possible future needs and directions to utilize CA for CO2 sequestration.

Activity enhancement of carbonic anhydrase in Chlamydomonas sp. for effective CO2 sequestration

ABSTRACT Present study describes the induction of carbonic anhydrase (CA) activity in Chlamydomon... more ABSTRACT Present study describes the induction of carbonic anhydrase (CA) activity in Chlamydomonas sp. in presence of calcium carbonate. Results indicated that calcium carbonate increased the activity of extracellular CA enzyme by 14-fold compared to culture grown at 3 % CO2, and by fourfold compared to the culture grown at 0.03 % CO2. CA was purified by gel exclusion followed by ion exchange chromatography. Molecular weight of the purified CA was found to be approximately 28 kDa. The enzyme showed optimum stability in the pH range of 8.0–9.0 and temperature of 35 °C. CA activity was inhibited with Hg2+ and Pb2+, while Zn2+ was found to accelerate its activity. Purified CA was employed for carbonation reaction with CO2 and the calcite produced was characterized by scanning electron microscope and X-ray diffraction. The enhancement in CA activity was found to be reliable, convenient, and may be employed for CO2 sequestration using whole cells or immobilized bioreactor systems.

Influence of mercury from fly ash on cattle reared nearby thermal power plant

The present investigation aims to assess the environmental and health impacts of fly ash containing mercury emitted from thermal power plant. The health effect of fly ash were studied using 20 lactating cattle reared within a 5-km radius of s thermal power plant for the possible effect of fly ash such as the alterations in hematological and biochemical parameters of blood, milk, and urine. Results indicated that the hemoglobin levels (6.65±0.40 g/dl) were significantly reduced in all the exposed animals. Biochemical parameters viz., blood urea nitrogen (27.35 ± 1.19 mg/dl), serum glutamate oxaloacetate transaminase (43.39±3.08 IU/l), albumin, and creatinine were found to be increased, whereas serum glutamate pyruvic transaminase (29.26±2.02) and Ca 2+ were observed to be statistically insignificant in exposed animals. Mercury concentrations estimated in the blood, milk, and urine of exposed (n020) and control (n020) animals were 7.41±0.86, 4.75±0.57, 2.08±0.18, and 1.05±0.07, 0.54±0.03, 0.20±0.02 μg/kg, respectively. The significant increase (P<0.01) in the levels of mercury in blood, milk, and urine of exposed animals in comparison to control indicated that the alterations of biochemical parameters in exposed cattle could be due to their long term exposure to fly ash mercury which may have direct or indirect impact on human populations via food chain.

In this study, an attempt was made to use micro-algal system for the production of biodiesel prec... more In this study, an attempt was made to use micro-algal system for the production of biodiesel precursors and simultaneous CO 2 mitigation. Chlorella sp. was found to have a higher growth rate as compared to the other algal species tested namely Chlamydomonas sp. and Synnecococcus sp. At different CO 2 concentrations (0.03%, 3%, 10% and 15%), the lipid productivity was 23.0, 20.0 and 27.3 mg/L/d respectively. Calcite produced was characterized using FT-IR, SEM and XRD. The FAME in crude biofuel was analyzed by GC-FID that found to contain palmitic acid (C16:0), docosapentaenoic acid (C22:5) and docosahexaenoic acid (C22:6). The calorific value of Chlorella sp. was found to be 29 kJ/g which is higher than values reported for fresh water microalgae making it a potential candidate to be used as an alternate fuel.

Biological CO 2 sequestration using algal reactors is one of the most promising and environmental... more Biological CO 2 sequestration using algal reactors is one of the most promising and environmentally benign technologies to sequester CO 2 . This research study was taken up to alleviate certain limitations associated with the technology such as low CO 2 sequestration efficiency and low biomass yields. The study demonstrates an increase in CO 2 sequestration efficiency by maneuvering chemically aided biological sequestration of CO 2 . Chlorella sp. and Spirulina platensis showed 46% and 39% mean fixation efficiency, respectively, at input CO 2 concentration of 10%. The effect of acetazolamide, a potent carbonic anhydrase inhibitor, on CO 2 sequestration efficiency was studied to demonstrate the role of carbonic anhydrase in calcite deposition. Calcite formed by both species was characterized by scanning electron microscopy coupled electron dispersive spectroscopy and X-ray diffraction. The overall scheme of calcite deposition coupled CO 2 fixation with commercially utilizable biomass as a product seems a viable option in the efforts to sequester increasing CO 2 emissions.
