The Ca2l-messenger system plays a crucial role in the regulation of steroid production in adrenal... more The Ca2l-messenger system plays a crucial role in the regulation of steroid production in adrenal zona-glomerulosa cells, as it is known to mediate the action of both angiotensin II and K+. In the present study we used intact isolated glomerulosa cells in which the cytosolic free Ca21 concentration ([Ca2+]') was clamped at various levels with the Ca2+ ionophore ionomycin in order to locate the site(s) of action of Ca2+. By measuring in parallel steroid synthesis and [Ca2+]0, we show that Ca2+ levels (50-860 nM) regulate the production of both pregnenolone (up to 669 + 71.1 % of the basal production) and aldosterone (up to 301 +42.2 %; EC50 = 303 nM). By contrast, Ca2+ did not stimulate the conversion of 11-deoxycorticosterone into aldosterone. Ca2+ modulation did not affect the formation of pregnenolone from freely diffusible analogues of cholesterol, indicating that

Macromolecular X-ray crystallography is routinely applied to understand biological processes at a... more Macromolecular X-ray crystallography is routinely applied to understand biological processes at a molecular level. However, significant time and effort are still required to solve and complete many of these structures because of the need for manual interpretation of complex numerical data using many software packages and the repeated use of interactive three-dimensional graphics. PHENIX has been developed to provide a comprehensive system for macromolecular crystallographic structure solution with an emphasis on the automation of all procedures. This has relied on the development of algorithms that minimize or eliminate subjective input, the development of algorithms that automate procedures that are traditionally performed by hand and, finally, the development of a framework that allows a tight integration between the algorithms.

Over the last decade we have witnessed the emergence of technologies such as libraries, Object Or... more Over the last decade we have witnessed the emergence of technologies such as libraries, Object Orientation, software architecture and visual programming. The common goal of these technologies is to achieve software reuse. Even though, many significant advances have been made in areas such as library design, domain analysis, metric of reuse and organization for reuse, there are still unresolved problems such as component inter-operability and framework design . We have investigated the use of interpreted languages to create a programmable, dynamic environment in which components can be tied together at a high level. This work has demonstrated the benefits of such an approach and has taught us about the features of the interpreted language that are key to a successful component integration.
