Increased nuclear protein O‐linked β‐N‐acetylglucosamine glycosylation (O‐GlcNAcylation) mediated... more Increased nuclear protein O‐linked β‐N‐acetylglucosamine glycosylation (O‐GlcNAcylation) mediated by high glucose treatment or the hyperglycemia of diabetes mellitus contributes to cardiac myocyte dysfunction. Mitochondrial proteins in cardiac myocytes are also submitted to O‐GlcNAcylation, however, whether excessive O‐GlcNAcylation alters mitochondrial function is incompletely studied. We have previously demonstrated decreased mitochondrial calcium concentration in cardiac myocytes exposed to high glucose. We hypothesize that excessive O‐GlcNAcylation may be responsible of this maladaptive phenomenon and that reducing excessive O‐GlcNAcylation may return mitochondrial calcium levels to normal. Our objective was to test this hypothesis, therefore, we incubated cultured neonatal mouse myocytes in high glucose (HG, 25mM) for 72 hours and inhibited O‐GlcNAcylation by adenoviral expression of a dominant negative mutant of the O‐GlcNAc transferase (dnOGT). Ischemic heart disease (IHD) is the single most common cause of death. New approaches to enhance ... more Ischemic heart disease (IHD) is the single most common cause of death. New approaches to enhance myocardial perfusion are needed to improve outcomes for patients with IHD. Thyroid hormones (TH) are known to increase blood flow; however, their usefulness for increasing perfusion in IHD is limited because TH accelerates heart rate, which can be detrimental. Therefore, selective activation of TH effects is desirable. We hypothesized that cell-type-specific TH receptor (TR) expression can increase TH action in the heart, while avoiding the negative consequences of TH treatment. We generated a binary transgenic (BTG) mouse that selectively expresses TRα1 in endothelial cells in a tetracycline-inducible fashion. In BTG mice, endothelial TRα1 protein expression was increased by twofold, which, in turn, increased coronary blood flow by 77%, coronary conductance by 60%, and coronary reserve by 47% compared with wild-type mice. Results: DRP1 is O-GlcNAcylated. Increased O-GlcNAcylation augments the level of the GTP-bound active form of DRP1 and induces translocation of DRP1 from cytoplasm to mitochondria. Conclusion: O-GlcNAcylation modulates DRP1 function, which has consequences for mitochondrial function. Significance: The modulation of DRP1 function by increased overall O-GlcNAcylation could play a significant role in the development of diabetic mitochondrial dysfunction.

Urocortin-2 (UCn2) peptide infusion increases cardiac function in patients with heart failure, bu... more Urocortin-2 (UCn2) peptide infusion increases cardiac function in patients with heart failure, but chronic peptide infusion is cumbersome, costly, and provides only short-term benefits. Gene transfer would circumvent these shortcomings. Here we ask whether a single intravenous injection of adeno-associated virus type 8 encoding murine urocortin-2 (AAV8.UCn2) could provide long-term elevation in plasma UCn2 levels and increased left ventricular (LV) function. Normal mice received AAV8.UCn2 (5 • 10 11 genome copies, intravenous). Plasma UCn2 increased 15-fold 6 weeks and > 11-fold 7 months after delivery. AAV8 DNA and UCn2 mRNA expression was persistent in LV and liver up to 7 months after a single intravenous injection of AAV8.UCn2. Physiological studies conducted both in situ and ex vivo showed increases in LV + dP/dt and in LV -dP/dt, findings that endured unchanged for 7 months. SERCA2a mRNA and protein expression was increased in LV samples and Ca 2 + transient studies showed an increased rate of Ca 2 + decline in cardiac myocytes from mice that had received UCn2 gene transfer. We conclude that a single intravenous injection of AAV8.UCn2 increases plasma UCn2 and increases LV systolic and diastolic function for at least 7 months. The simplicity of intravenous injection of a long-term expression vector encoding a gene with paracrine activity to increase cardiac function is a potentially attractive strategy in clinical settings. Future studies will determine the usefulness of this approach in the treatment of heart failure.

To examine whether excessive protein O -GlcNAcylation plays a role in the dysfunction of the diab... more To examine whether excessive protein O -GlcNAcylation plays a role in the dysfunction of the diabetic heart, we delivered adenovirus expressing O -GlcNAcase (Adv-GCA) into the myocardium of STZ-induced diabetic mice. Our results indicated that excessive cellular O -GlcNAcylation exists in the diabetic heart, and that in vivo GCA overexpression reduces overall cellular O -GlcNAcylation. Myocytes isolated from diabetic hearts receiving Adv-GCA exhibited improved calcium transients with a significantly shortened T decay ( P <0.01) and increased sarcoplasmic reticulum Ca 2+ load ( P <0.01). These myocytes also demonstrated improved contractility including a significant increase in +dL/dt and −dL/dt and greater fractional shortening as measured by edge detection ( P <0.01). In isolated perfused hearts, developed pressure and −dP/dt were significantly improved in diabetic hearts receiving Adv-GCA ( P <0.05). Here we have characterized the functional effects of urocortin 2 on mouse cardiomyocytes and elucidated the underlying signalling pathways and mechanisms. Mouse ventricular myocytes were field-stimulated at 0.5 Hz at room temperature. Fractional shortening and [Ca 2+ ]i transients were measured by an edge detection and epifluorescence system respectively. Western blots were carried out on myocyte extracts with antibodies against total phospholamban (PLN) and PLN phosphorylated at serine-16. Urocortin 2 elicited time-and concentration-dependent positive inotropic and lusitropic effects (EC50: 19 nM) that were abolished by antisauvagine-30 (10 nM, n = 6), a specific antagonist of corticotrophin releasing factor (CRF) CRF2 receptors. Urocortin 2 (100 nM) increased the amplitude and decreased the time constant of decay of the underlying [Ca 2+ ]i transients. Urocortin 2 also increased PLN phosphorylation at serine-16. H89 (2 mM) or KT5720 (1 mM), two inhibitors of protein kinase A (PKA), as well as KN93 (1 mM), an inhibitor of Ca 2+ /calmodulin-dependent protein kinase II (CaMKII), suppressed the urocortin 2 effects on shortening and [Ca 2+ ]i transients. In addition, urocortin 2 also elicited arrhythmogenic events consisting of extra cell shortenings and extra [Ca 2+ ]i increases in diastole. Urocortin 2-induced arrhythmogenic events were significantly reduced in cells pretreated with KT5720 or KN93.

Cardiac fibroblasts (CF) express adenosine (ADO) receptors, and pharmacological evidence suggests... more Cardiac fibroblasts (CF) express adenosine (ADO) receptors, and pharmacological evidence suggests the possible involvement of the A2(A2aand A2b) receptor (A2aR and A2bR) subtypes in inhibiting cell functions involved in fibrosis. The main objective of this study was to define the contributions of A2aand/or A2breceptors in modulating ADO-induced decreases in CF functions. For this purpose, CF were either treated pharmacologically or had the A2aR or A2bR levels modified through the use of recombinant adenovirus or siRNA. The assessment of mRNA expression in adult rat CF yielded evidence for A1R, A2bR, A2aR, and A3R. Endogenously or exogenously enhanced ADO significantly inhibits CF proliferation, collagen, and protein synthesis. A2R and A2aR agonists, although capable of inhibiting CF protein and collagen synthesis, were unable to define the contributions derived from A2aR or A2bR.
