To investigate the anti-tumor activity of GT-enriched extract of galla rhois (GEGR) against lung carcinomas, alterations in the cytotoxicity, apoptosis activation, cell cycle progression, migration ability, tumor growth, histopathological structure, and the regulation of signaling pathways were analyzed in Lewis lung carcinoma (LLC1) cells and LLC1 tumor bearing C57BL/6NKorl mice, after exposure to GEGR. A high concentration of GT (69%) and DPPH scavenging activity (IC50=7.922 µg/ml) was obtained in GEGR. Nevertheless, a genetic transformation procedure for this cultivar has not yet been reported. Meanwhile, various genome editing technologies such as transfection of clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated (Cas) ribonucleoprotein complexes into cells are in the limelight. Utilizing the Cas9-expressing genotype possessing a reference genome can simplify the verification of an off-target effect, resolve the economic cost of Cas9 endonuclease preparation, and avoid the complex assembly process together with single-guide RNA (sgRNA) in the transfection approach. Thus, this study was designed to generate Cas9-expressing 'Heinz 1706' lines by establishing an Agrobacterium tumefaciens-mediated transformation (ATMT) procedure. Here, we report a rapid and reproducible transformation procedure for 'Heinz 1706' by finetuning various factors: A. tumefaciens strain, pre-culture and co-culture durations, a proper combination of phytohormones at each step, supplementation of acetosyringone, and shooting/rooting method. Particularly, through eluding subculture and simultaneously inducing shoot elongation and rooting from leaf cluster, we achieved a short duration of three months for recovering the transgenic plants expressing Cas9. The presence of the Cas9 gene and its stable expression were confirmed by PCR and qRT-PCR analyses, and the Cas9 gene integrated into the T 0 plant genome was stably transmitted to T 1 progeny. Therefore, we anticipate that our procedure appears to ease the conventional ATMT in 'Heinz 1706', and the created Cas9-expressing 'Heinz 1706' lines are ultimately useful in gene editing via unilateral transfection of sgRNA into the protoplasts.

The level of mRNA, inactive form (InACF) and active form (ACF) for C3 showed a regular pattern, which they were higher in the FVB/N and C57BL/6N mice than C3H/HeN mice. However, the level of C3b fragments (C3bα and β) derived from C3 protein were constantly maintained in the liver of FVB/N, C3H/HeN and C57BL/6N mice in spite of the strain difference on the transcriptional and translation level of C3. Here, we compared the substitution activity and the substitution range of two base-editing systems, APOBEC1-nCas9 and nCas9-PmCDA1, in the protoplasts of Glycine max, Brassica napus, and Nicotiana tabacum. To prevent unwanted nucleotide substitution, we manipulated the length of guide RNA and found the change of nucleotide substitution activity in the target window of nCAS9-PmCDA1. Based on these results, the specific C to T conversion in the acetolactate synthase gene of N. tabacum was induced to generate herbicide-resistant plants. During the screening of herbicide-resistant plants, we found that ubiquitin promoter-driven base-editor system was much efficient than 35S promoter-driven base-editor system. This study provides guidelines on which a base editor to use and describes how to fine-tune a guide RNA for precise substitutions in plants.
