Milk and dairy products represent potential nutritive environment for the growth of some pathogen... more Milk and dairy products represent potential nutritive environment for the growth of some pathogen bacteria such as Salmonella spp., Escherichia coli, Staphylococcus aureus. Our study aimed to assess the incidence of pathogen bacteria through classical and molecular methods in order to perform a risk analysis in Transylvanian production area. The material was represented by 160 ripened dairy products from the following categories: soft, medium-hard and hard. We have isolated E.coli in 121 samples (75.62%), which represent a concerning number. All the samples were confirmed by PCR method targeting the stress protein A gene sequence. No positive samples were found for Salmonella spp. and Staphylococcus aureus. At the risk analysis performed there was a medium level of probability and gravity for consumer contamination with pathogenic E.coli by consumption of dairy products. It was concluded that further studies are needed in order to assess the toxicity of the E.coli strains isolated from the dairy products investigated so as to accurately assess the risk for contamination. A more rigorous hygiene during the milking and manufacturing of dairy products is recommend, especially within the traditional systems.

The slaughter process in its current form is responsible to numerous physiological impacts on the... more The slaughter process in its current form is responsible to numerous physiological impacts on the animal and as a consequence affects the quality of the meat which is produced. The kosher slaughter is performed by a Rabbi (a certified Jewish religious priest) employed by the meat processing facility. The aim was to compare different types of slaughter methods and restraining techniques in order to determine the positive and negative sides of each method and to assess the hygienic quality of the meat produced from this method of slaughter. The information regarding kosher slaughter was gathered from one slaughterhouse located in Israel, Haifa city, and specific aspects related to the conventional slaughter technology, was acquired from a slaughterhouse located in Maramures County, where we critically observed the cattle slaughter technology, with focus on stunning and bleeding steps. In order to compare the hygienic quality of kosher beef with conventional beef the research material consisted by 40 samples of beef, half of the samples were represented by kosher beef and the other half (conventional beef) were used as control. Total viable and Enterobacteriaceae count were performed from the collected samples according with standards methods. In case of conventional slaughter, with respecting of good hygienic practices (GHP), bleeding will not open the esophagus and trachea, preventing thus the possibility of contamination. Stunning performed in conventional slaughter could increase the risk of contamination of carcasses with prions in case of penetration of the skull. In case of kosher meat, the results regarding total plate count revealed values between 2.78±0.6 log cfu/ cm 2 and 3.40±0.2 log cfu/cm 2 . In case of Enterobacteriaceae germs, we noticed that 3 samples shown a negative results. For the rest of the samples, the microbial load ranged between 2.54±0.62 log fcu/cm 2 and 3.1±0.32 log fcu/cm 2 . In comparison, the conventional meat (control), present an elevated total microbial load, ranged between 4.77±0.56 log cfu/cm 2 and 5.34±0.21 log cfu/cm 2 . The Enterobacteriacee load ranged between 4.1±0.56 log cfu/cm 2 and 5.33±0.2 log cfu/cm 2 . Ritual kosher slaughter is a humane way of sacrificing animals for human consumption although there are many parameters which need to be carefully monitored in order to guarantee the welfare of the slaughtered animals. And thus, inherently this method is prone for animal welfare violation. Microbiological risk regarding total plate and Enterobacteriaceae count in case of kosher compared with conventional meat is low.

The aim was to reveal the prevalence of post-mortem lesions in two large-scale bovine slaughterho... more The aim was to reveal the prevalence of post-mortem lesions in two large-scale bovine slaughterhouses found in Romania so as to point out the potential hazards for human health. The proper examination of animal carcasses and organs that are destined for human consumption is essential in order to ensure food safety but not all the time properly made due to lack of time during the process of slaughter. The results showed that the most frequent lesions found within the examined organs were caused by parasitic infestation (Echinococcus granulosus; Dyctiocaulus viviparus) and the highest prevalence was found in the lungs. The pathology of lesions found in bovine organs is very variable in the slaughterhouse, being able to report strange aspects such an ectopic liver in the lung parenchyma. Measures should be taken in the farms due to this high prevalence of parasites which causes both health problems as well as economical losses.

Concerning Increase in Antimicrobial Resistance Patterns of Pathogenic Strains of Salmonella Isolated in Poultry Meat Products

Salmonella is considered to be one of the major foodborne pathogens associated with the consumpti... more Salmonella is considered to be one of the major foodborne pathogens associated with the consumption of contaminated poultry meat products. To the best of our knowledge this is the first extended research performed on a number of Salmonella strains isolated during 2011-2021 from poultry meat products in Romania. The aim of this study was to characterize the prevalence of pathogenic Salmonella serovars, antimicrobial susceptibility, and antimicrobial resistance genes in 112 Salmonella isolates recovered from raw poultry meat products. The results showed that Salmonella enterica serovars Enteritidis and Typhimurium were the common serotypes (56%; 25%). Overall, the majority of the isolates were resistant to at least three tested antimicrobials. High resistance was observed for tetracycline (84%), nalidixic acid (78%), and ampicillin (78%) in pathogenic Salmonella isolated during the period 2016-2021. All the pathogenic Salmonella isolated during 2016-2021 tested positive to at least one resistance gene encoding for tetracycline resistance, with the tetA gene being the most prevalent (62%). In addition, 64% (24/37) of the Salmonella isolates carried at least one of the genes (blaCMY-2, blaSHV1, blaTEM1) that code for β-Lactams resistance. The findings in this study showed a high prevalence of multi-drug resistant (MDR) Salmonella serovars in poultry meat products and a concerning increase of resistance patterns. The continuous occurrence of more resistant strains implies that effective measures should be strictly applied in this particular food chain in order to prevent their spread and guarantee microbial safety.

The aim was to evaluate the quality aspects of raw meat destined for processing in fast-food unit... more The aim was to evaluate the quality aspects of raw meat destined for processing in fast-food units and to compare it with that of meat obtained in the traditional systems. Our study revealed that the water holding capacity (WHC) in poultry meat obtained in the industrial systems and used for processing in fast-foods is very variable and also very high compared to the one obtained in the traditional systems. When examining sensorially the quality aspects we found that the colour of the poultry meat (legs) is whiter, paler and in the breast areas we noticed a pathology known the "wooden breast". In the case of pork meat, we obtained similar results, the industrially obtained meat showing a lighter colour and lower consistency. The protein percentage was correlated with the low quality of the meat obtained in the industrial system. Raw meat destined for processing in the fastfood units should be improved given the lower quality revealed. Measures should be taken also in the production units so as to prevent the pathological lesions found, such as "wooden breast" in poultry meat which we found to be very often observed.

Lipids oxidation represents one major cause of sensorial changes and quality loss of meat. Also, is responsible for reducing the shelf life, being influenced by the temperature and time of meat storage. The aim of this study was represented by the assessment of oxidation spoilage processes through determination of thiobarbituric acid (TBARS) correlated with the load and configuration of psychotropic microflora of beef, pork, poultry and fish meat. The research material analysed was represented by 27 samples collected from 3 different batches of beef, pork, and poultry and fish meat, purchased from the retail network of a hypermarket from Cluj County. The samples have been carried in isothermal bags at the Food inspection and control laboratory from FVM Cluj-Napoca, and they have been kept at freezing temperatures (-20°C). In order to assess in dynamic the oxidative spoilage processes, there have been carried out 3 analyses during the experiment: July 2014, December 2014 and May 2015. From the research of the give results regarding the oxidative processes it has been determined in the meat samples that the thiobarbituric acid (TBARS) represented in dynamics an ascendant evolution, the highest values being in poultry in the first day of the experiment 14.4±0.4 and 19.74±1.16 nmol/100 g malondialdehyde (MDA). Rephrase, difficult to follow The microbial load of meat samples presented a descendent evolution during the storage at -20ºC, from 4.9±0.31 log cfu/g at the beginning of the experiment to 4.1±0.26 log cfu/g in the last day.

An Analysis of <i>E</i> <i>scherichia coli</i> Isolations for Antimicrobial Resistance Genes

This research was conducted in order to reveal antimicrobial resistance patterns and the prevalen... more This research was conducted in order to reveal antimicrobial resistance patterns and the prevalence of genes encoding resistance in Escherichia coli isolated from raw poultry meat.One hundred samples of poultry meat were collected, and 51 isolates of E. coli were recovered and further tested for antimicrobial susceptibility to 11 antimicrobials, using the classical disk diffusion method. Fifteen isolates, showing a high degree of resistance among the collection were examined for antimicrobial resistance genes by multiplex polymerase chain reaction.Resistance was most frequently observed to tetracycline (84.26%), ampicillin (73.03%) and sulfamethoxazole (66.29%). The tetracycline resistance genes TetA and TetB and beta‐lactam TEM gene were frequently detected in the tested isolates. Genes responsible for resistance to sulfonamides (33.33%) and trimethoprim (26.66%) were also found.E. coli isolates recovered from raw poultry meat are highly resistant to antimicrobials and carry antimicrobial resistance genes that could be transferred to other microbes in the food chain, even important human pathogens.Practical ApplicationsThe findings from this work provide baseline data for further antimicrobial resistance studies especially in our country, were antimicrobial resistance epidemiology needs constant improvement.To our knowledge, this is one of the first reports regarding the identification of various resistance genes identified in E. coli meat isolations in Romania. Moreover, the work contributes to the risk assessment concerning the possible E. coli threat in food products and draws a warning signal for further actions that can assist the implementation of efficient measures to prevent possible adverse public health consequences.

In this study we aimed to assess the efficiency of organic acids in different concentrations rega... more In this study we aimed to assess the efficiency of organic acids in different concentrations regarding pathogens as Salmonella, Listeria and Escherichia on beef, which can cause food borne illness in humans. The samples were sterilized using UV radiation for 30 minutes, afterwards being contaminated with 1 ml of microbial suspension (0.5 MacFarland). We used reference bacterial strains for Salmonella Enteritidis, Escherichia coli and Listeria monocytogenes. The samples were subjected to decontamination procedure by introducing 25mL of solution of lactic, acetic or citric acid in concentration of 1%, 2% and 3%. The results showed a reduction of initial pathogen load, ranging from 0.32 to 7.78 log CFU/g, depending on the type of acid, concentration and pathogen sensitivity. After decontamination, standardized methods have been used for the isolation of pathogenic germs. Based on statistical analysis we conclude that pathogens have a different sensitivity to the action of acid solutions, their sensitivity in ascending order being: Listeria monocytogenes, Salmonella Enteritidis and Escherichia coli. Among the organic acids, the most efficient was lactic acid, followed by acetic acid and less efficient citric acid. The greatest reduction of germs was determined by the concentration of 3%.

Antimicrobial abuse and chaotic administration is a common fact in poultry production leading to the emergence, selection and spreading of drug resistant E.coli in both veterinary and human medicine. This research was conducted on a number of 41 E.coli isolates from poultry meat (n=28) and feacal material from humans (n=12). The resistance genes of the main classes of antibiotics (TetA, TetB, dfrAI, sulI, TEM) were evaluated through PCR multiplex. It was revealed a high prevalence of the multi-resistant strains in E.coli strains isolated from poultry meat. None of the strains isolated proved positive for stx1, stx2, genes that are characteristic for Shiga toxigenic E.coli which causes infections in humans. Still, the PCRs revealed that tetA and tetB (tetracycline resistance genes) were the most prevalent genes (64.5%) found in poultry isolates while in human cases (60.8%, dfrA17 (52.4%) (trimethoprim resistance genes) and sulI (41.3) (sulfonamide resistance genes) proved the most frequent. Even though the resistance patterns in the E.coli strains isolated from poultry meat and the ones isolated from humans are not similar, still there is a high prevalence of human contamination with resistant E.coli by poultry meat consumption. The resistance genes studied have been isolated in a higher percent than in other studies made in European countries, which leads us to the conclusion that E.coli in this region is highly mutagenic and can be consider a reservoir of resistance genes.

The prevalence of Sarcocystis spp. in pork and wild boar sample from Transylvania area has not be... more The prevalence of Sarcocystis spp. in pork and wild boar sample from Transylvania area has not been extensively studied and many consumers are not aware of the possible risks that they are exposed to. This study was conducted on a total of 150 samples, of which 120 were represented by diaphragmatic pillars collected from domestic pigs raised in extensive systems and 30 by muscle tissue from wild boars. The method used for identifying the Sarcocystis spp. cysts was represented by classical trichinelloscopy, which normally can also detect these forms. From the total number of samples examined, 64% were found positive and 23% declared with massive contamination of the carcass. In conclusion we established that the pork meat produced in the traditional system represents a risk in consumers' contamination with Sarcocystis spp.

Residual antimicrobial effect of week organic acids on spoilage psychrotrophs at pig carcasses

The aim of this researchwas to assess the residual antimicrobial effect of 3% lactic and acetic a... more The aim of this researchwas to assess the residual antimicrobial effect of 3% lactic and acetic acid solutions regarding the load and configuration of psychrotrophs at porcine carcasses. During October 2016 and December 2016, 18 pork meat samples were collected from a comercial slaughterhouse in Transylvania. Collected samples were sprayed with 3% acetic acid and lactic acid solutions by spraying on the surface of meat samples. Each sample was divided into three sub-samples, from which two were treated with 3% organic acids solutions and one was the control sample. Experimental design were carried out over a 14day period with microbial analyses at day 0, 1, 5, 9, 14. After spraying with organic acid solutions, the samples were kept at 2-4°C for 24 hours, and the following microbiological determinations were carried out: total load of psychrotrophic germs and isolation of microorganisms from the genera Pseudomonas, Aeromonas, Yersinia and Enterobacteriaceae family. The most sensitive psychrophic bacteria regarding the decontamination effect of lactic and acetic acid were Aeromonas spp. and Yersinia spp., both species being completely inhibited after 24 hours since application. For all microbiological criteria analyzed, lactic and acetic acid shown an obvious residual antimicrobial effect during the shelf life of pork carcasses, when compared with control samples (p<0.05).

The globalization of the food chain has led nowadays to the increase of the risks related to the ... more The globalization of the food chain has led nowadays to the increase of the risks related to the dissemination of food poisoning episodes which extend over international borders. Due to this fact, risk analysis of each step of the production chain "from farm to fork" has become necessary. The specific aim of this study was to perform risk analysis in two swine slaughtering units and to highlight the microbiological hazards taking into account the performed analyses. The technological steps that present important risks for meat hygiene state and could jeopardize the production were the refrigeration step and the storage step. In the small capacity slaughterhouse "B", there was a severe operational deficiency noticed, consisting of the inaccurate removal of the tonsils, which can lead to a high risk of microbial contamination. During the evaluation of the probability for the risk of occuring, in the big capacity slaughterhouse there was a low estimated probability (0-10%) compared to unit "B" where the probability was high (>80%), mainly due to inaccurate removal of the tonsils. During evaluation of risk exposure, with the help of representative scales, marked with colours according to severity, there was a high tolerance noticed in the case of unit "B" represented with the red colour. Following the way of communication and report making, both units presented deficienciesin establishing the alert mechanisms.

Comparative Study on the Fat Acids Profile from Buffalo Milk as Quality and Traceability Markers

The nutritional aspects have begun to receive an increasing attention, particularly the short-cha... more The nutritional aspects have begun to receive an increasing attention, particularly the short-chained ones, polyunsaturated, cis and trans. The conjugated fatty acids are potentially involved as positive or negative factors for human health (Wolff,1995; Williams, 2000). This study focuses on the compositional investigation of the fatty acids from buffalo milk, as quality and traceability markers making also a comparison with the cow milk. After the analysis made we established that the main fatty acid is the oleic one, in a percentage of 24,29% compared to the cow milk were its value is even higher 33,5%. In the case of the linoleic acid the percentage was again higher in the case of the cow milk samples (2,4%) compared to the buffalo milk (1,87). The situation is reversed at the linolenic acid, where we revealed a higher value in the case of the buffalo milk samples. The percentage of polyunsaturated fatty acids is higher in the cow milk compared to buffalo milk.

Assessment of Small-scale Buffalo Milk Dairy Production-A Premise for a Durable Development

Buffalo husbandry is an important source of income for a number of small-scale producers in Roman... more Buffalo husbandry is an important source of income for a number of small-scale producers in Romania that is why an assessment of its' product's quality is much needed for improvement and evaluation of their vulnerability to international competition. In order to ascertain possible developments in the buffalo dairy sector and to broadly identify areas of intervention that favor small-scale dairy producers, the study examined the potential to improve buffalo milk production by evaluating its authenticity and hygienic quality. The methods used involved the molecular testing (PCR-technique) for identifying cow, sheep or goat DNA in the dairy products' samples collected from the small-scale producers market. The hygienic quality of these samples was determined through classical microbiology methods, highly developed techniques (Trek System) and PCR for bacterial species confirmation. The results showed that a high percent (65%), from the products found were adulterated with other species milk, mostly cow milk. The most commonly falsified buffalo dairy products were the cheese and the traditional product "telemea". The prevalence of the bacterial species identified belonged to Listeria innocua and Listeria welshmeri. The conclusion of this study is the need of a durable development system in this particular dairy chain to improve and assure the authenticity and quality of the small-scale producers' products and their reliability for the consumers.

The present study proposed the cheese differentiation, according to geographical production area ... more The present study proposed the cheese differentiation, according to geographical production area and with respect to species (cow, sheep) of two traditional cheese specialties, (salty and ripened), produced in Transylvania, Romania. For this purpose, the elemental profile and carbon isotopic ratios ( 13 C/ 12 C) of cheese and extracted casein were corroborated through statistic supervised techniques to get the best discrimination markers. The manganese content, along with Rare Earth Elements (REE) concentrations, proved to be very powerful predictors, for the traditional salted cheese mainly, due to the direct influence of the local salted water. Despite that proposed techniques are not acknowledged methods for species differentiation, this approach allowed a successful discrimination of the animal species that produced the raw materials for cheese manufacturing (milk). The results generated by the developed chemometric model, for species differentiation, were compared with those obtained using Isoelectric focusing (IEF) and DNA tests. The proposed association of isotopic and elemental markers allowed a differentiation better than 92 % for geographical provenance, of each investigated cheese specialties while, for species discrimination (cow vs. sheep) a percentage of 100 % was obtained.

The study focused on the incidence of enterotoxigenic Escherichia coli (ETEC) and verotoxigenic E... more The study focused on the incidence of enterotoxigenic Escherichia coli (ETEC) and verotoxigenic E. coli (VTEC) in raw milk and traditional dairy cheeses marketed in Romania, characterizing the virulence and antibiotic resistance genes of these isolates. One hundred and twenty samples of raw milk and 80 samples of unpasteurized telemy cheese were collected and cultured according to the international standard protocol. All the characteristic E. coli cultures were analyzed for the presence of STa, STb, LT, stx1, and stx2 toxicity genes. The ETEC/VTEC strains were tested for the presence of antibiotic resistance genes, such as aadA1, tetA, tetB, tetC, tetG, dfrA1, qnrA, aaC, sul1, bla SHV , bla CMY , bla TEM , and ere(A), using PCR. The results showed that 27 samples (18.62%) were positive for one of the virulence genes investigated. 48.1% (n = 13) tested positive at the genes encoding for tetracycline resistance, tetA being the most prevalent one (61.5%; n = 8). A high percent (33.3%; n = 9) revealed the beta-lactamase (bla TEM ) resistance gene, and none of the samples tested positive for bla CMY and bla SHV genes. The genes responsible for resistance to sulfonamides (sul1) and trimethoprim (dfrA1) were detected in rates of 14.8% (n = 4) and 7.4% (n = 2), respectively. E. coli is highly prevalent in raw milk and unpasteurized cheeses marketed in Romania. These strains might represent an important reservoir of resistance genes which can easily spread into other European countries, given the unique market.

The computerized modelling systems represent an intelligent predictive way for the individual's s... more The computerized modelling systems represent an intelligent predictive way for the individual's state of health, through the assessment of the effects of the particular risk factors needed in the development of a model used in the prevention of cardiac diseases. The main purpose was to perform correlations between the socio-economic and clinical factors in order to observe food impact on cardiovascular diseases and to obtain a modelling system that would prevent cardiac diseases. In order to develop on-line applications to calculate the cardiac risk score, modern technologies have been developed and applied which allow -through Web instruments -access to the available information: anytime, anywhere secured and differentiated according to the category of users. Furthermore, the risk calculation is adjusted according to some variables taken into account -the social, economic and health parameters. The computerized system uses the forms to extract certain features and to enable health prediction. The originality of the application consists in the development of a new variable, namely the health clinical factors, which contribute to accurately calculate the risk factor, differently from other calculating systems which rely only on the health indices. The system that prevents cardiac diseases is constructed as a reliable predictive model with the purpose of achieving a rapid screening in order to reduce the morbidity and mortality due to cardiovascular diseases.
