The entomopathogenic nematode (EPN) Steinernema australe was isolated from Isla Santa Magdalena i... more The entomopathogenic nematode (EPN) Steinernema australe was isolated from Isla Santa Magdalena in Chile and identified as a good alternative for controlling Aegorhinus nodipennis (Coleoptera: Curculionidae) larvae. This weevil is native to the south of Chile and some regions in Argentina, causing the decline and ultimate death of plants in berry orchards. The major problem brought about by the weevil is caused by the larvae, which spend between nine and eleven months below ground, feeding inside the roots of the plants. This study seeks to increase S. australe’s efficacy through an artificial selection process using an odor stimulus. We selected infective juveniles (IJs) that followed the stimulus in order to reach larvae at a depth of 30 cm to achieve this objective. Larvae infected with selected IJs and IJs from the original stock were compared under laboratory, greenhouse, and field conditions. Distribution and ecology of some species of red algae (Rhodophyta) in the Ebro River Basin The im... more Distribution and ecology of some species of red algae (Rhodophyta) in the Ebro River Basin The implementation of the Water Framework Directive (2000/60/CE) in the Ebro River Basin and the evaluation of the ecological status of rivers, lakes and wetlands are increasing the knowledge about the distribution and ecology of aquatic organisms, including the red algae (Rhodophyta). In this work we show new data concerning the distribution, habitat and environmental ranges of five species of Rhodophyta in the Ebro River Basin with scarce records: Bangia atropurpurea, Chroodactylon ornatum, Chroothece rupestris, Compsopogon coeruleus and Thorea hispida. The former three species showed an optimum development in calcareous mountain streams while the latter two species did in lower reaches where salinity and eutrophication increase.

Lehenik, ardo-hondakinetatik polifenolak eta gantz-azidoak, bakoitza bere aldetik, erauzi genituen jariakin gainkritikoak erabiliz. Polifenolak ez dira oso egonkorrak, ordea, eta gorputzean bioeskuragarritasun baxua dute. Arazo horiek konpontzeko, polifenolak mikrokapsularatzeko teknika bat garatu genuen. Bigarren zutabean, etorkizun hurbilean kannabisa sendagai bezala erabili ahal izateko, 3 urrats eman genituen. Alde batetik, landare ezberdinak efektu ezberdinekin lotzea ahalbide dezakeen landareen kannabinoideen hatz-marka osatzeko analisi-teknika garatu genuen. Bestetik, gernuan eta plasman kannabinoideak eta euren metabolitoak kuantifikatzeko metodo bat ere garatu genuen. Azkenik, konposatu puruen eta landare mota jakinen produkzioa optimizatzeko, kimiotipo ezberdineko landareen hazkuntzan zeharreko kannabinoideen eta terpenoen garapena aztertu genuen. Pulsed light (PL) is a non-thermal technology able to inactivate enzymes. This work aimed to assess the potential inactivation of PME by PL and to explain how it occurs. To this, a suspension of the enzyme was treated with PL up to 128 J/cm 2. The inactivation curve was built and structural changes were determined by far-UV circular dichroism, intrinsic and extrinsic fluorescence and spectrophotometry. Furthermore, the presence/absence of unfolding intermediaries was assessed by phase-diagram analysis. PL was able to inactivate PME with a first-order kinetic rate of 0.034 cm 2 /J. The inactive protein seemed to have a molten-globule structure, due to the conserved secondary structure and modified tertiary structure. Phase diagram data fitted a single line, which is consistent with absence of unfolding intermediaries. A significant increase in absorbance at 420 nm (p<0.05) showed that PL promoted enzyme aggregation.

Las numerosas infraestructuras destinadas a paliar los efectos de inundaciones y sequías en la De... more Las numerosas infraestructuras destinadas a paliar los efectos de inundaciones y sequías en la Demarcación Hidrográfica del Segura hacen de esta la más regulada de Europa. Presas, canales, sistemas de riego y otras obras hidráulicas provocan un deterioro en la corriente fluvial, que presenta a menudo consecuencias irreversibles y, en ocasiones, desconocidas. El presente trabajo es el resultado del interés por analizar el impacto que genera la presa de un embalse en la dinámica fluvial. De esta forma, se pretende evaluar el grado de naturalidad hidrogeomorfológica de un tramo del río Segura comprendido entre el embalse de la Fuensanta y la desembocadura del río Tabilla. Para ello, se ha empleado el Índice Hidrogeomorfólogico (IHG), el cual ha sido desarrollado con éxito anteriormente en la Demarcación Hidrográfica del Ebro.

Lipoxygenase (LOX) causes enzymatic rancidity in some foods. This study aimed to determine the structural changes associated with the PL inactivation of LOX and used measurements of residual activity, temperature, spectropolarimetry, fluorescence, spectrophotometry, free sulfhydryl and carbonyl contents and electrophoresis. LOX inactivation was non-loglinear, with a slight temperature rise. PL significantly increased the concentration of carbonlys and free-sulfhydryls of LOX and caused loss of ellipticity and intrinsic fluorescence, a red shift in the intrinsic fluorescence peak and an increase in 1-anilino-8naphthalenesulfonate fluorescence. The turbidity of LOX sample increased during inactivation and electrophoretic bands faded. Phase diagram analysis showed no evidence of formation of intermediates. In summary, the inactivation of LOX by PL followed a Weibull kinetics, is exclusively photochemical and is an all-or-none process where the protein gets oxidized, decreases its α-helix content, unfolds and aggregates.

This study investigated the effect of pulsed light on the activity of polygalacturonase, and on the structure of the enzyme by means of fluorescence emission spectra, free sulphydryl detection and analysis of changes in parameter A and phase-diagram. The results showed that pulsed light is able to inactivate polygalacturonase in buffer, with > 90 % reduction of enzyme activity after applying 128 J/cm 2 and a first-order kinetic constant of 0.0426 cm 2 /J under the experimental conditions. The free sulfhydryl detection revealed the rupture of sulfhydryl bridges. Fluorescence spectra analysis showed that the tertiary structure of polygalacturonase was changed. Phase diagram analysis shows the existence of only two populated states. It is suggested that the inactivation of polygalacturonase by pulsed light is an all-or-none process where disulfide bridges are broken and the enzyme is unfolded.

Since the 1980s, research into entomopathogenic nematodes (EPNs) in Latin America has produced ma... more Since the 1980s, research into entomopathogenic nematodes (EPNs) in Latin America has produced many remarkable discoveries. In fact, 16 out of the 117 recognized species of EPNs have been recovered and described in the subcontinent, with many more endemic species and/or strains remaining to be discovered and identified. In addition, from an applied perspective, numerous technological innovations have been accomplished in relation to their implementation in biocontrol. EPNs have been evaluated against over 170 species of agricultural and urban insects, mites, and plant-parasitic nematodes under laboratory and field conditions. While much success has been recorded, many accomplishments remain obscure, due to their publication in non-English journals, thesis dissertations, conference proceedings, and other non-readily available sources. The present review provides a brief history of EPNs in Latin America, including current findings and future perspectives.

Pulsed light (PL) is a non-thermal method of food preservation that is able to inactivate PPO. The aim of this work was to gain insight into the mechanism of PPO inactivation by PL. To this, the kinetics of PPO inactivation by PL was measured, together with associated changes in tryptophan fluorescence, KI fluorescence quenching and turbidity; and results were analysed by parameter A and phase diagram methods. Enzyme inactivation followed the Weibull model. Tryptophan fluorescence decreased during PL treatment, as well as the parameter A, while Stern-Volmer constants increased and turbidity was constant. The phase diagram showed only two populated states. There was a high correlation between the loss of activity and parameter A. Results indicate that under the experimental conditions, the inactivation of PPO by PL is an all-or-none process where the enzyme progressively unfolds with no evidence of aggregation.

This dataset gathers information about the macroinvertatebrate samples and environmental variable... more This dataset gathers information about the macroinvertatebrate samples and environmental variables collected on rivers of the Ebro River Basin (NE Iberian Peninsula), the second largest catchment in the Iberian Peninsula. The collection is composed of 1,776 sampling events carried out between 2005 and 2015 at more than 400 sampling sites. This dataset is part of a monitoring network set up by the Ebro Hydrographic Confederation, the official body entrusted with the care of the basin, to fulfill the requirements of the European Water Framework Directive. Biological indices based on the freshwater macroinvertebrate communities were used to evaluate the ecological status of the water bodies within the basin. Samples were qualitatively screened for all occurring taxa. Then, all individuals from all taxa in a quantitative subsample of each sample were counted. Experimental studies in animals infer the development of persistent infection in the kidneys. In humans, recent studies suggest the possibility of persistent renal infection and chronic kidney disease. Considering the discrepancies between published studies on viral RNA detection in urine of convalescing WNV-positive patients, we explored the use of electron microscopy (EM) with anti-WNV E protein antibody immunogold labeling to detect virus in the urine sediment from a subset of study participants in the Houston WNV cohort. In 42% of study participants, we evaluated who had visible sediment present in urine after centrifugation; viral particles consistent with the size and morphology of WNV were successfully detected using EM in the urine sediment up to 9 years postinfection. Pulsed light (PL) is a food processing technology initially intended for microbial inactivation t... more Pulsed light (PL) is a food processing technology initially intended for microbial inactivation that can potentially be applied in other UV-light based processes. PL lamps emit high intensity broad spectrum light from UV to infrared. This research tested the use of PL as part of an advanced oxidation process (AOP) for degrading polluting dyes. Experiments were performed in a batch reactor, and the efficiency of the AOP under different parametric values: dye concentration, pH and H 2 O 2 doses on Direct yellow 106 decolourization was followed by spectrophotometry. Effects on chemical oxygen demand and electrical efficiency were also determined. Decolourization process follows a pseudo-first order kinetic. It was improved by increasing H 2 O 2 concentrations and low pH; while there was little influence of dye concentration, perhaps due to the reactor configuration. The highest constant rate observed was 0.0410 cm 2 / J. The decolourization by PL/H 2 O 2 fits to a typical mechanism of a conventional UV/H 2 O 2 process; since no direct photolysis or thermal effects were observed. Mineralization was incomplete likely due to the highly resonant structure of the dye. An electrical energy per order of 1009 kWh/m 3 /order was calculated. Ninety % of colour removal was reached at 40 J/cm 2 , which is achievable in less than 30 s by most of PL systems existing in the market. The PL/H 2 O 2 process seems to be useful for decolourization of wastewater and could offer the advantage of a very fast degradation.

The hydrolysis process was studied to ensure complete hydrolysis of glucuronide conjugates and the extraction of a total amount of analytes. Initially, urine and plasma blank samples were spiked with THC-COOH-glucuronide and THC-glucuronide, and four different pretreatment methods were compared: hydrolysis-free method, enzymatic hydrolysis with Escherichia Coli β-glucuronidase, alkaline hydrolysis with 10 M NaOH, and enzyme-alkaline tandem hydrolysis. The evolution of major cannabinoids and terpenes during the growth of Cannabis sativa plants was ... more The evolution of major cannabinoids and terpenes during the growth of Cannabis sativa plants was studied. In this work, seven different plants were selected: three each from chemotypes I and III and one from chemotype II. Fifty clones of each mother plant were grown indoors under controlled conditions. Every week, three plants from each variety were cut and dried, and the leaves and flowers were analyzed separately. Eight major cannabinoids were analyzed via HPLC-DAD, and 28 terpenes were quantified using GC-FID and verified via GC-MS. The chemotypes of the plants, as defined by the tetrahydrocannabinolic acid/ cannabidiolic acid (THCA/CBDA) ratio, were clear from the beginning and stable during growth. The concentrations of the major cannabinoids and terpenes were determined, and different patterns were found among the chemotypes. In particular, the plants from chemotypes II and III needed more time to reach peak production of THCA, CBDA, and monoterpenes. Differences in the cannabigerolic acid development among the different chemotypes and between monoterpene and sesquiterpene evolution patterns were also observed. Plants of different chemotypes were clearly differentiated by their terpene content, and characteristic terpenes of each chemotype were identified.

The tumor demonstrated little or no immunoreactivity for a broad panel of antibodies yet strong, diffuse immunoreactivity with CD68. On electron microscopy, characteristic rectangular to rhomboid crystalline inclusions were not present. Electron-dense granules resembling peroxisomes were present, sometimes in association with elongated granular structures having a periodic, lattice-like arrangement. Metastatic ASPS was confirmed by demonstration of an ASPSCR1-TFE3 fusion and imaging studies that excluded metastatic Xp11.2 translocation renal cell carcinoma. The primary site was subsequently identified in the lower extremity.

High performance liquid chromatography tandem mass spectrometry (HPLC-MS/MS) has been successfull... more High performance liquid chromatography tandem mass spectrometry (HPLC-MS/MS) has been successfully applied to cannabis plant extracts in order to identify cannabinoid compounds after their quantitative isolation by means of supercritical fluid extraction (SFE). MS conditions were optimized by means of a central composite design (CCD) approach, and the analysis method was fully validated. Six major cannabinoids [tetrahydrocannabinolic acid (THCA), tetrahydrocannabinol (THC), cannabidiol (CBD), tetrahydrocannabivarin (THCV), cannabigerol (CBG), and cannabinol (CBN)] were quantified (RSD < 10%), and seven more cannabinoids were identified and verified by means of a liquid chromatograph coupled to a quadrupole-time-of-flight (Q-ToF) detector. Finally, based on the distribution of the analyzed cannabinoids in 30 Cannabis sativa L. plant varieties and the principal component analysis (PCA) of the resulting data, a clear difference was observed between outdoor and indoor grown plants, which was attributed to a higher concentration of THC, CBN, and CBD in outdoor grown plants.

In this study, supercritical fluid extraction has been successfully applied to a sequential fract... more In this study, supercritical fluid extraction has been successfully applied to a sequential fractionation of fatty acids and polyphenols from wine wastes (2 different vitis vinifera grapes). To this aim, in a 1st step just fatty acids were extracted and in a 2nd one the polyphenols. The variables that affected to the extraction efficiency were separately optimized in both steps following an experimental design approach. The effect of extraction temperature flow, pressure, and time were thoroughly evaluated for the extraction of fatty acids, whereas the addition of methanol was also considered in the case of the polyphenols extraction. A quantitative extraction with high efficiency was achieved at a very short time and low temperatures. Concerning quantification, fatty acids were determined by means of gas chromatography coupled to mass spectrometry after a derivatization step, whereas the polyphenols were analyzed by means of high performance liquid chromatography coupled to tandem mass spectrometry and the Folin-Ciocalteu method.
