Results.-On 16 patients, only 10 questionnaires were usable (questionnaires not filled, evaluation before, after not possible). At the end of 3 weeks, pain is very clearly improved with P 0.013, the score of kinesiophobia also (31.8 vs 41.2) with P in 0.009. The scores of acceptance are improved 27.44 against 21.88 at the exit but not significant with P in 0.14. The scores of change of behavior PSCOQ are all improved for all the patients for the hillside preservation (will to improve its strategies to face) P 0.0057 and no change for the intention. There was also a significant improvement of the anxiety (P in 0.03), not significant for the depression (P in 0.1). The physical parameters (schober, outstrip fingers ground, poplités angles, outstrip heel spank, shirado, sorensen) were improved. Conclusion.-For all the patients, is turned out an increase of the implication of the patient in its care, at the end of the program. It also seems that this type of program brings an improvement of the acceptance of the pain chronic without it is significant on our small population as well as of the capacity to manage better their disease. Studies on a bigger population and a remote follow-up would be desirable.

Les lesions de l'ADN causees par 1 O 2 ainsi que leurs consequences biologiques ont ete etudi... more Les lesions de l'ADN causees par 1 O 2 ainsi que leurs consequences biologiques ont ete etudiees. Pour cela une source chimique ayant la propriete de produire uniquement de l'oxygene moleculaire singulet ( 1 O 2 ), durant la thermodissociation d'un endoperoxyde soluble dans l'eau, le 3,3'-(1,4-naphthylidene)dipropionate (NDPO 2 ), sans intermediaires reactifs ou produits secondaires, a ete utilisee. L'analyse de la mobilite electrophoretique de l'ADN simple brin ou double brin sur gel d'agarose, a permis de mesurer des coupures franches simples et doubles brin lors du traitement de l'ADN avec 1 O 2 genere par NDPO 2 . La prevention de ces lesions de l'ADN (coupures franches simples) par des biomolecules a ete recherchee. Les polyamines, spermine et spermidine, se sont montrees efficaces. Les consequences letales et mutagenes de ces lesions ont ete demontrees apres introduction des plasmides dans des bacteries ou cellules de mammiferes. Une importante augmentation de la frequence de mutation en fonction de la concentration du NDPO 2 a ete observee dans les plasmides de simple et double brin apres introduction dans des cellules COS7 de singe. Ceci suggere l'existence d'un processus de reparation d'ADN endommage par 1 O 2 qui produit des erreurs dans les cellules de mammiferes. Le spectre de mutation revele que 98% des mutations impliquent les paires de base G :C. La haute capacite mutagene est attribuee au grand nombre de lesions des guanines de l'ADN. Cela suggere que les residus de guanine sont la cible de 1 O 2 et que ces lesions causent directement les mutations.

The generation of electronically excited molecular oxygen 1O2 has been shown to occur in several ... more The generation of electronically excited molecular oxygen 1O2 has been shown to occur in several biological systems, such as photooxidation of a variety of biological compounds and xenobiotics ("photodynamic action") and also enzymatic reactions. The high reactivity of 1O2 with unsaturated compounds, sulfides and amino groups arises from its electrophilicity and relatively long lifetime. Thus, biological targets for 1O2 having the above functional groups include unsaturated fatty acids, proteins, enzymes and DNA. There is interest in the role of nutrition in the prevention and pathogenesis of cancer. Epidemiological studies in humans have suggested that carotenoids aid in cancer prevention. Lycopene and oxycarotenoids are present at significant levels in cells and plasma. Extensively conjugated biomolecules such as carotenoids act largely on physical quenching of 1O2 and in much lesser extent on chemical reaction. In this study we observed the protective effect of beta-carotene and lycopene entrapped in human albumin (HSA) against the oxidative 1O2 attack of 2'-deoxyguanosine (dGuo). Photosensitization with methylene blue associated with Chelex resine or Polymer-Rose bengal (Sensitox) and thermodecomposition of water-soluble endoperoxide 3,3'-(1,4-naphthylidene)dipropionate were employed to generate 1O2. The detection of 8-oxo-7,8-dihydro-2'-deoxyguanosine(8-oxodGuo) and 4-hydroxy-8-oxo-7,8-dihydro-2'-deoxyguanosine(4-OH-8-oxodGuo) were performed using reversed phase HPLC with UV, electrochemical detection and by electrospray ionization mass spectrometry. Results showed a significant decrease in the amount of 8-oxodGuo in the presence of lycopene. The percentages of 4-OH-8-oxodGuo and 8-oxodGuo measured were 50% and 70% lower than the control, respectively. These data indicate that carotenoids entrapped in albumin can be an efficient quencher of 1O2 and may be of interest in protecting against the deleterious effect of this excited state molecule.

These potentially deleterious reactions are controlled by a system of enzymatic and nonenzymatic antioxidants which eliminate prooxidants and scavenge free radicals. The ability of the lipid-soluble carotenoids to quench singlet molecular oxygen may explain some anticancer properties of the carotenoids, independent of their provitamin A activity. Tocopherols are the most abundant and efficient scavengers of hydroperoxyl radicals in biological membranes. Water-soluble antioxidants include ascorbate and cellular thiols. Glutathione is an important substrate for enzymatic antioxidant functions and is capable of nonenzymatic radical scavenging. Thiols associated with membrane proteins may also be important to the antioxidant systems. Interactions between the thiols, tocopherols, and other compounds enhance the effectiveness of cellular antioxidant defense.
