Esta revisión resume los modelos teóricos de interacciones parásito-hospedero que se encuentran d... more Esta revisión resume los modelos teóricos de interacciones parásito-hospedero que se encuentran disponibles en la literatura científica. Se revisaron publicaciones internacionales en revistas especializadas sobre parasitología y ecología. Estudios teóricos y prácticos de las interacciones parásitohospedero, en algunos casos, sugieren variaciones evolutivas y ecológicas. La evolución de las interacciones parásitohospedero no se limita a consideraciones coevolutivas, sino que tiene efectos directos sobre caracteres de historia de vida del hospedero, evolución del sexo, comportamiento reproductivo, selección sexual e, incluso, sobre transferencia génica. El estudio de las interacciones parásito-hospedero, como fuerzas modeladoras de la evolución y ecología de los organismos, tiene gran importancia, pues aunque, actualmente, carece de evidencia firme en la práctica en algunas de sus áreas y permanece como difícil de demostrar, es clara de manera teórica.

The Blueprint to Safeguard Europe’s Water Resources will set out the Commission’s views on the policy initiatives that will be most appropriate to address these problems. This report sets out the problems that the EU faces in achieving effective water management and develops and assesses a list of potential options to tackle these problems.

The results suggest that approximately 37 different proteins are expressed in the presence of Zn 2+ , which also downregulates the protein and transcriptional levels of TvCP65. The result is a negative effect on trichomonal cytotoxicity. The differentially expressed proteins were identified by mass spectrometry analysis.

Proteomic characterization of the subpellicular cytoskeleton of Toxoplasma gondii tachyzoites

Toxoplasma, the causative agent of toxoplasmosis in animals and humans, has a subpellicular cytos... more Toxoplasma, the causative agent of toxoplasmosis in animals and humans, has a subpellicular cytoskeleton that is involved in motility, cell shape and invasion. Knowledge of components of the cytoskeleton is necessary to understand the invasion mechanisms as well as for the identification of possible therapeutic targets. To date, most cytoskeletal components of Toxoplasma remain unidentified due mainly to the lack of reproducible methods for their isolation. Based on the successful isolation of the cytoskeleton, it was possible to report for the first time, the proteomic characterization of the subpellicular cytoskeleton of Toxoplasma formed by 95 cytoskeletal proteins through proteomic analysis by tandem mass spectrometry of one dimension SDS PAGE. By bioinformatic analysis of the data, proteins were classified as: 18 conventional cytoskeletal proteins; 10 inner membrane complex proteins, including 7 with alveolin repeats; 5 new proteins with alveolin like repeats; 37 proteins associated with other organelles and 25 novel proteins of unknown function. One of the alveolin like proteins not previously described in Toxoplasma named TgArticulin was partially characterized with a specific monoclonal antibody. Presence of TgArticulin was exclusively associated with the cytoskeleton fraction with a cortical distribution. Functions for the several molecules identified are proposed. This manuscript describes, for the first time, the proteome of the subpellicular cytoskeleton of Toxoplasma gondii. The importance of this study is related to the role of the cytoskeleton in the highly invasive capability of a parasite that causes abortion, blindness, and death by encephalitis in immunocompromised patients. Proteomic characterization of the cytoskeleton of T. gondii tachyzoites was possible by the development of a successful procedure for the isolation of the subpellicular cytoskeleton. Knowledge of the composition of the cytoskeleton of Toxoplasma is fundamental for the understanding of the motility and host cell invasion mechanisms, and for the future design and development of toxoplasmicidal drugs with effects against specific components of the cytoskeleton of this parasite that are absent in mammal host cells.

Catalysis and Stability of Triosephosphate Isomerase from Trypanosoma brucei with Different Residues at Position 14 of the Dimer Interface. Characterization of a Catalytically Competent Monomeric Enzyme

In homodimeric triosephosphate isomerase from Trypanosoma brucei (TbTIM), cysteine 14 of each the... more In homodimeric triosephosphate isomerase from Trypanosoma brucei (TbTIM), cysteine 14 of each the two subunits forms part of the dimer interface. This residue is central for the catalysis and stability of TbTIM. Cys14 was changed to the other 19 amino acids to determine the characteristics that the residue must have to yield catalytically competent stable enzymes. C14A, C14S, C14P, C14T, and C14V TbTIMs were essentially wild type in activity and stability. Mutants with Asn, Arg, and Gly had low activities and stabilities. The other mutants had less than 1% of the activity of TbTIM. One of the latter enzymes (C14F) was purified to homogeneity. Size exclusion chromatography and equilibrium sedimentation studies showed that C14F TbTIM is a monomer, with a k(cat) approximately 1000 times lower and a K(m) approximately 6 times higher than those of TbTIM. In C14F TbTIM, the ratio of the elimination (methylglyoxal and phosphate formation) to isomerization reactions was higher than in TbTIM. Its secondary structure was very similar to that of TbTIM; however, the quantum yield of its aromatic residues was lower. The analysis of the data with the 19 mutants showed that to yield enzymes similar to the wild type, the residue must have low polarity and a van der Waals volume between 65 and 110 A(3). The results with C14F TbTIM illustrate that the secondary structure of TbTIM can be formed in the absence of intersubunit contacts, and that it has sufficient tertiary structure to support catalysis.
